Interaction of cytokinin and abscisic acid in growth regulation in Arabidopsis thaliana

Abstract:

Kyselina abscisová a cytokininy jsou dva rostlinné hormony, které se často objevují v současném výzkumu odpovědi rostlin na stres. Aby se přizpůsobily stresu, vyvinuli rostliny řadu regulačních mechanismů. Korelace mezi hladinami několika fytohormonů a jejich signalizačními sítěmi hraje klíčovou roli v adaptaci rostlin na stres. Od objevení antagonistického vztahu mezi kyselinou abscisovou a cytokininovou signalizací uběhla dlouhá doba, ale základní mechanismus není zdaleka jasný. Tato práce byla zaměřena na detailní studium vlivu interakce mezi cytokininy a kyselinou abscisovou na růst a vývoj rostlin Arabidopsis thaliana. Základem této práce bylo zkoumání mutantní linie aba1-1 na genetickém pozadí Ler-0, která má poruchu v biosyntetickém genu kyseliny abscisové ABA1. Ukázalo se, že exogenním přídavkem kyseliny abscisové bylo možné revertovat účinek dané mutace. Po ošetření mutantních rostlin exogenně přidanými cytokininy byly pozorovány výrazné morfologické rozdíly. Proto následovala analýza fotosyntetických parametrů rostlin, která odhalila, že cytokininy narušují fotosyntézu více než kyselina abscisová. Sledování vlivu cytokininů a kyseliny abscisové na hladinu karotenoidů ukázalo snížení jejich hladin a změny v NPQ (nefotochemickém zhášení). To bylo předpokladem pro studium exprese biosyntetických genů, která ukázala, že cytokininy a kyselina abscisová reagují nejenom prostřednictvím svých signalizačních prvků, ale taky na úrovni biosyntézy.

Abstract:

Abscisic acid and cytokinins are two plant hormones that frequently appear in current research on plant responses to stress. To adapt to stress, plants have developed several regulatory mechanisms. The correlation between the levels of several phytohormones and their signaling networks plays a key role in the adaptation of plants to stress. A long time has passed since the discovery of the antagonistic relationship between abscisic acid and cytokinin signaling, but the underlying mechanism is far from clear. This work was focused on a detailed study of the effect of the interaction between cytokinins and abscisic acid on the growth and development of Arabidopsis thaliana plants. The basis of this work was the investigation of the mutant line aba1-1 on the genetic background Ler-0, which has a disorder in the biosynthetic gene abscisic acid ABA1. It turned out that the exogenous addition of abscisic acid made it possible to reverse the effect of the mutation. Significant morphological differences were observed after the treatment of mutant plants with exogenously added cytokinins. Analysis of the photosynthetic parameters revealed that cytokinins disrupt photosynthesis more than abscisic acid. The determination of pigments proved a negative impact of cytokinins and ABA on carotenoid level accompanied by changes in NPQ (non-photochemical quenching). Further, the expression of selected genes in carotenoid metabolism revealed that cytokinins and abscisic acid interact on the level of carotenoid and ABA biosynthesis.
