Factor VIII inhibitors in Hemophilia A

Abstract:

Tato bakalářská práce pojednává o původu a účinku inhibitorů proti koagulačnímu faktoru VIII, které jsou častou komplikací u pacientů s těžkou hemofilií A. Vznik těchto protilátek je podmíněn nejen typem mutace způsobujícím hemofilii A u daného pacienta, ale také geny pro imunitní odpověď v kombinaci s dalšími negenetickými faktory, jako je typ léčebných prostředků podávaných pacientovi. Práce dále shrnuje laboratorní a molekulární diagnostiku a také základní léčebné strategie pro minimalizaci účinků inhibitorů.

Abstract:

This thesis investigates the cause and effect of inhibitors against the coagulation factor VIII which are a frequent complication in patients with severe hemophilia. The presence of these antibodies depends not only the mutation type causing hemophilia A in the patient, but is also influenced by several genes responsible for the imunne response as well as a plethora of enviromental factors such as treatment strategy used in the patient. The thesis further summarises the laboratory and molecular diagnosis as well as essential treatment strategies used in such patient to minimise the effect of the inhibitors.
