Abstract:

Disertace je předložena jako soubor čtyř prací: (i) V první práci je ukázáno využití hmotnostního spektrometru vybaveného analyzátorem doby letu využívajícího k ionizaci laserem za přítomnosti matrice (MALDI TOF MS) k rozpoznávání tkání. Pro rozpoznání bylo použito tkání myšího organismu zdravého a stiženého nedostatkem enzymu L-alanin:glyoxalát transferázy způsobující hyperoxalurii prvního typu. Dobrá reprodukovatelnost výsledků byla zajištěna přídavkem zlatých nanočástic během procesu depozice běžných matric na povrch tkání. Signály naměřených hmotnostních spekter byly podrobeny analýze hlavních komponent (PCA) a neuronovým sítím (ANN). Obě metody byly schopny správně rozlišit původ tkání. (ii) Instrumentace MALDI TOF MS byla aplikována pro studium procesu pulzní laserové depozice chalkogenidových skel o různém složení prvků (Ge2Sb2Te5 a As-S-Se). Byla provedena analýza jak materiálů vstupujícího do procesu (PLD), tak výsledných deponovaných tenkých vrstev. Pro většinu signálů hmotnostních spekter bylo nalezeno stechiometrické složení odpovídajícího ionizovaného klastru. U systému As-S-Se byl popsán dopad změny stechiometrického složení chalkogenidového skla na výsledná hmotnostní spektra. (iii) Byly sledovány produkty reakcí pyridiniumbetainu kyseliny chlorodithiofosforečné se šesti adamantanovými deriváty obsahujícími aminoskupinu. Produkty reakcí byly charakterizovány fyzikálně-chemickými metodami nukleární magnetické resonance, infračervenou a Ramanovou spektrometrií, rentgenovou krystalografií, hmotnostní spektrometrií. Celkové složení produktů C, H, N bylo stanoveno pomocí elementární analýzy. Použitím instrumentace LDI TOF MS bylo možno potvrdit stechiometrii u některých předpokládaných reakčních produktů.

Abstract:

Dissertation contains in total four works: (i) In the first work the use of a mass spectrometer equipped with a time of flight detector and using the process of matrix assisted laser desorption ionization (MALDI TOF MS) to recognize the status of tissue was shown. As a demonstration, a mouse organism suffering from a deficiency of L-alanine:glyoxylate amino- transferase causing hyperoxaluria 1 disease was used. Good result reproducibility was provided by the addition of gold nanoparticles during the common matrix deposition process onto the tissue surface. Mass spectrometric signals were subjected to principal component (PCA) and artificial neural network analysis (ANN). Both methods were able to distinguish the tissue origin correctly. (ii) Instrumentation of MALDI TOF MS was applied to study the process of pulsed laser deposition of chalcogenide glass with different compositions of its elements (Ge2Sb2Te5 and As-S-Se). Analysis of the input materials (for PLD), and also the resulting thin layers was done. The stoichiometric composition corresponding to ionized clusters was determined for most of the signals present in the mass spectra. The change of the chalcogenide glass stoichiometry in the mass spectra was described for the system As-S-Se. (iii) The reaction products of chlorodithiophosphoric acid pyridiniumbetaine with six adamantane derivatives containing amino groups were examined. The reaction products were characterized using physicochemical methods such as nuclear magnetic resonance, infrared and Raman spectrometry, Roentgen crystallography and mass spectrometry. The content of C, H, N was determined by elemental analysis. It was possible to confirm the stoichiometry of several supposed reaction products using LDI TOF MS instrumentation.
