Functional characterisation of LDL receptor gene variants

Abstract:

Diplomová práce je věnována studiu patogenních variant v genu pro LDL (low density lipoprotein) receptor, které jsou nejčastější příčinou familiární hypercholesterolemie (FH). Jedná se o autozomálně dominantní poruchu. Některé sekvenční varianty genu LDLR (low density lipoprotein receptor) mají vliv na nesprávné sbalení proteinu, který je následně akumulován v endoplazmatickém retikulu a není schopen vychytávat LDL částice na cytoplazmatické membráně buněk. Pro stanovení diagnózy FH je zásadní znalost vlivu konkrétních sekvenčních variant na strukturu a funkci LDLR proteinu. V praktické části bylo analyzováno 10 sekvenčních variant LDLR genu. Cílem práce bylo ověřit možnost využití metod kvantitativní analýzy pro funkční charakterizaci LDL receptoru. Konfokální laserová mikroskopie živých buněk byla doplněna o kvantitativní analýzu průtokovou cytometrií. U všech analyzovaných variant byla ovlivněna funkce LDLR proteinu. Některé sekvenční varianty způsobují zadržení LDLR na endoplazmatickém retikulu, jiné pravděpodobně ovlivňují recyklaci LDL receptoru v buňce.

Abstract:

This thesis is devoted to the study of pathogenic variants in LDL (low density lipoprotein) receptor gene. These variants are the most common cause of familial hypercholesterolemia (FH). It is an autosomal dominant disorder. Some sequence variants of gene LDLR (low density lipoprotein receptor) affect the improper protein folding which is subsequently accumulated in the endoplasmic reticulum and unable to capture LDL particles on the plasma membrane of cells. To determine the diagnosis of FH, a knowledge of the impact of specific sequence variants on the structure and function of the LDLR protein is essential. In the experimental part, 10 sequence variants of the LDLR gene were analyzed. The aim of the thesis was to verify the possibility of using the methods of quantitative analysis for the functional characterisation of the LDL receptor. Live cell imaging microscopy was supplemented by quantitative analysis by flow cytometry. The function of the LDLR protein was affected in all analyzed variants. Some sequence variants cause a retention of LDLR protein on the endoplasmic reticulum, others may influence LDL receptor recycling.
