Detection of electrons in microscopy and modification of detectors for an analysis the specific biological structures

Abstract:

V této diplomové práci se budeme zabývat elektronovou mikroskopií, nejprve popíšeme problematiku elektronové mikroskopie (s důrazem na optimální detekci elektronů a modifikace detektorů pro analýzu specifických biologických preparátů), včetně metod přípravy preparátů a nastíníme si základní charakteristiku biomembrán. V kapitole „metodika“ popíšeme parametry mikroskopu, na němž probíhalo měření. V této kapitole bude též popsán preparát, který byl připraven klasickou chemickou cestou. Budeme používat různé detektory (detektor sekundárních elektronů, detektor zpětně odražených elektronů, detektor prošlých elektronů, detektor sekundárních elektronů v nízkém vakuu) s cílem získání co nejlepšího výstupního signálu. S ohledem na to, že preparáty jsou biologické vzorky, bylo nutno značně modifikovat pracovní nastavení mikroskopu pro každý preparát zvlášť. Preparáty byly většinou připraveny metodou freeze-etching (mrazové leptání) a pozorovány skenovacím elektronovým mikroskopem. Jednalo se o buňky lidské promyeloidní leukémie (HL-60), u kterých jsme se zaměřili hlavně na membrány. Tyto buňky byly současně pozorovány na transmisním elektronovém mikroskopu, což umožnilo srovnání obou metod snímání.

Abstract:

In this thesis, we will deal with the electron microscopy. Firstly, we will describe the issue of the electron microscopy (with the emphasis on the optimal detection of electrons and modifications of detectors for the specific biological sample analysis) including the methods of the specimen preparation and we will outline the basic characteristics of biomembranes. In the chapter „methodology“, we will describe the parameters of the microscope, which the measurement was enacted with. In this chapter, the preparation will also be described, which was prepared by the classic chemical way. We will use different detectors (a secondary electron detector, a backscattered electron detector, a transmitted electron detector and a low vacuum secondary electron detector) with the aim of getting the best output signal. In view of the fact that the preparations are biological samples, it was necessary to modify operational setting of the microscope for each preparation separately. The preparations were mostly prepared by the freeze-etching method and observed by the electrone scanning microscope. The preparations were cells of the human leukemia (HL-60) and we focused mainly to their membranes. These cells were simultaneously observed with the transmissive electrone microscope, which enabled the comparison of both methods of scanning.
