Utilization of the Ames fluctuation test

Abstract:

Bakalářská práce se zabývá detekcí mutagenních účinků standardních látek a povrchových vod pomocí Amesova fluktuačního testu, který je modifikovanou verzí klasického Amesova testu. Pro test byly použity mutantní bakteriální kmeny Salmonella typhimurium (TA 98, TA 100). V teoretické části práce jsou charakterizovány složky životního prostředí a jejich kontaminace, jednotlivé testy genotoxicity, charakteristika mutace, mutageneze a s tím související karcinogeneze, vlastnosti mutagenních látek a jejich vliv na organismus. Cílem praktické části bylo seznámit se s metodikou Amesova fluktuačního testu a jeho použití při hodnocení vzorků standardních látek a povrchových vod.

Abstract:

The bachelor thesis deal with detection of mutagenicity effect of standard mutagens and surface water by means of Ames fluctuation test, which is a modified version of the classical Ames test. The assay uses a bacterial mutant strain of Salmonella typhimurium TA 98 and TA 100. In the teoretical part are charakterized the environmental contamination of the environment, described the tests of mutagenicity, characteristics of mutation, mutagenesis, carcinogenesis and mutagenic substances and their effect on organism. The aim of the practial part was take up with methodics of Ames fluctuation test and its use in evaluating the samples of surface water and standard mutagens.
