Cytotoxicity of Different Polyaniline Forms

Abstract:

Cílem diplomové práce je stanovit cytotoxicitu přečištěných polyanilinových prášků. Jako přečišťovací média budou použita metanol a kyselina chlorovodíková. Práce navazuje na dříve publikované studie zabývající se cytotoxicitou polyanilinu ve formě prášku a jeho přečištěním. Polyanilin je zkoumán hlavně pro svou schopnost vést elektrický proud. Poly-anilin se vyskytuje ve třech různých formách, ale jenom jedna je stabilní a to semi-oxidovaná forma, emeraldine. Emeraldine se dále vyskytuje ve dvou formách a to 1) zeleně zbarvená, vodivá forma polyanilinu emeraldinová sůl, jejíž vodivost se pohybuje v řádech 100 Sm-1 a 2) emeraldinová báze, což je nevodivá forma polyanilinu. Emeraldinovou sůl lze převést na emeraldinovou bázi pomocí změny pH. Vyhodnocení bude probíhat na extraktech připravených z předem syntetizovaného a přečištěného polyanilinového prášku. Metody vyhodnocení budou MTT test, který je vhodný pro stanovení viability buněk a průtoková cytometrie, která je vhodná k rozeznání typu buněčné smrti.

Abstract:

The aim of my thesis is to determine cytotoxicity of purified polyaniline powders. The me-thanol and hydrochloric acid will be chosen as the purification media. The cytotoxicity of the standard polyaniline has been already determined and published. The thesis is therefore a follow up study based on the results of those studies. Polyaniline is examined mainly for its conductivity. The polyaniline exists in three diferent forms, but only one is stable. The stable form of polyaniline is semi-oxidated and is called emeraldine. Emeraldine further exists in two forms, and that is 1) green coloured, conducted form of polyaniline emer-aldine salt, whose conductivity is in the range of 100 Sm-1 and 2) emeraldine base, which is nonconductive form of polyaniline. The emeraldine salt could be transferred to emeraldine base by the change of pH. The evaluation of cytotoxicity will be determined on the ex-tracts of polyaniline powder. The polyaniline powder will be synthesized and modified. The evaluation methods will be MTT test, which is suitable for determination of viability of cells and flow cytometry, which is suitable for identifying the type of cell death.
