Interactions of telomeric proteins

Abstract:

The telomeres of eukaryotic chromosomes are created by repetitive DNA sequences and associated protein complexes. These protein complexes form functional units. The main function of telomeres is to protect of genetic information against non-complete DNA replication and to maintain stability of genome. The significant complex of telomere proteins is called shelterin. Shelterin consists of six protein subunits: TRF1, TRF2, TIN2, POT1, Rap1 and TPP1. The binding of shelterin proteins to telomeric DNA modulates the formation of specific loop-like structures. These loop-like structures are used to detect and distinguish telomeres from DNA double strand breaks and are important for regulation of telomere length. This thesis summarized today’s knowledge of interactions between shelterin proteins and their function in telomere length regulation with emphasis on structure and function of protein TIN2. Protein TIN2, the central protein of shelterin complex, was partially purified. In experimental part of this thesis the purification method of TIN2 using of ion exchange chromatography is described.

Abstract:

Teloméry eukaryotických chromozómov sú tvorené opakujúcimi sa nukleotidovými sekvenciami DNA a naviazanými proteínovými komplexy. Tieto proteínové komplexy tvoria na telomérach funkčné celky. Hlavnou funkciou telomér je zabrániť stratám genetickej informácie pri neúplnej replikácii DNA a zaistiť stabilitu genómu. Významným komplexom telomérových proteínov je komplex nazývaný shelterín. Shelterín pozostáva zo šiestich proteínov: TRF1, TRF2, POT1, TPP1, TIN2 a Rap1. Väzba shelterínových proteínov na telomérovú DNA moduluje tvorbu špecifických slučkovitých štruktúr. Tieto slučkovité štruktúry zabezpečujú odlíšenie telomér od dvojreťazcových zlomov DNA a sú dôležité pre reguláciu dĺžky telomér. V tejto práci boli zhrnuté súčasné poznatky o interakciách proteínov shelterínu a ich funkcii v regulácii dĺžky telomér s dôrazom na štruktúru a funkciu proteínu TIN2. Proteín TIN2, ktorý je centrálnym proteínom shelterínového komplexu, bol v rámci experimentálnej časti čiastočne purifikovaný. Pre túto purifikáciu bola optimalizovaná metóda iontomeničovej chromatografie (IEC).
