Abstract:

Tato bakalářská práce se zabývá významem stanovení protilátek proti antigenu DFS-70 u pacientů, u nichž byla metodou nepřímé imunofluorescence prokázána přítomnost antinukleárních protilátek. Protilátky anti-DFS70 reprezentují potenciálně důležitý biomarker, který může být klinicky využit k vyloučení diagnózy systémového onemocnění pojiva. Nevýhodou nepřímé imunofluorescence je však její nízká specifita pro tento typ onemocnění, což limituje přesnou diagnózu. Je tedy doporučováno potvrdit přítomnost protilátek anti-DFS70 dalšími metodami. V této práci jsem provedla stanovení protilátek anti-DFS70 metodou ELISA a imunoblot. Vzájemný vztah těchto metod jsem zhodnotila na základě korelace naměřených výsledků. Následně jsem porovnala počty pozitivit získaných těmito metodami s počtem pozitivních výsledků získaných prostřednictvím nepřímé imunofluorescence. Mezi metodami ELISA a imunoblot byla prokázána velmi silná kladná korelace. Dále bylo zjištěno, že četnost pozitivit získaných metodou ELISA je ve velmi těsné shodě s počtem pozitivit získaných nepřímou imunofluorescencí. Pro potvrzení přítomnosti protilátek anti-DFS70 bych se tedy přikláněla k testování metodou ELISA.

Abstract:

This bachelor thesis deals with the significance of determination of anti-DFS70 antibodies in ANA-positive patients captured by IIF testing. Anti-DFS70 antibodies represent a potentially important biomarker that can be used to exclude the diagnosis of systemic autoimmune rheumatic diseases. However, the disadvantage of IIF is its low specificity for this type of disease, which limits the accurate diagnosis. Therefore, it is recommended to confirm the presence of these antibodies by other methods. In this work, I performed the determination of anti-DFS70 antibodies by ELISA and immunoblot. I evaluated the conformity of these methods based on the correlation of the measured results. Subsequently, I compared the number of positives obtained by these methods with the number of positives obtained through the IIF. Between ELISA and immunoblot was found a very strong positive correlation. Also, it was found that the frequency of positives obtained by ELISA is in very close conformity with the number of positives obtained by IIF. Therefore to confirm the presence of anti-DFS70 antibodies, I would be inclined to ELISA.
