Telomeres as an indicator of relative age and aging of organism in birds

Abstract:

Telomery jsou považovány za jeden z faktorů souvisejících se stárnutím a bývají označovány jako markery biologického věku. Nacházejí se na koncích eukaryotických chromozomů, kde chrání genetickou informaci před poškozením. Jejich délka se s každým mitotickým dělením buňky zkracuje, čímž ztrácí svoji funkci. V současné době existují mnohé molekulární metody měřící délky telomer a vykazující patřičné rozdíly v jejich délce v závislosti na čase. Dobrou modelovou skupinou pro studium telomer jsou ptáci, kteří obývají rozmanitá prostředí a mají různé životní strategie. U většiny druhů ptáků se telomery s přibývajícím věkem zkracují, ale existují výjimky, například buřňáček dlouhokřídlý (Oceanodroma leucorhoa). Telomery jsou proto často studovány jako faktory spojené se stárnutím, jelikož u mnoha druhů včetně člověka byla prokázána jejich souvislost s délkou života a s faktory, které na ni působí. Cílem této práce bylo 1) provést literární rešerši o molekulárních metodách používaných na měření telomer a 2) srovnat změnu délky telomer u jedinců různých věkových kategorií dlouhodobě pozorovaných druhů ptáků.

Abstract:

Telomeres are considered to be one of the factors related with ageing and are labelled as the marker of biological age. They are located at the ends of eukaryotic chromosomes where they protect genetic information from damage. However, telomere length is shortened with every mitotic division which causes its loss of function. Currently, many molecular methods exist to measure the length of telomeres clearly showingtheir length differences in the course of time. Birds represent a good model group for the studies on telomeres, because they inhabit various environments and have different life strategies. Telomere length of most of the bird species is shortening with age, but theexceptions exist, e.g. in Leach's storm petrel (Oceanodroma leucorhoa). Considering this, the telomeres are often studied as the predictors related to ageing process, as their connection with the life span and other factors was proved in many species including human . The main aims of this thesis were 1) to write a reviewon molecular methods used for the telomeres measurement and 2) to compare the change in telomeres length of the individuals with different age from long term studies of various bird species.
