Determination of representative fusarium mycotoxins using LC-MS

Abstract:

Tato diplomová práce se zabývá studiem mykotoxinů neboli studiem toxických látek, které jsou produkované vláknitými mikroskopickými houbami rodu Fusarium. Teoretická část je zaměřena na obecné seznámení s rozsáhlou skupinou mykotoxinů, klasifikací jednotlivých toxických látek spolu s nejčastějšími producenty, dále také jejich možnými toxickými účinky, stručným charakteristickým popisem nejvýznamnějších fusariových mykotoxinů a jejich případným povoleným množstvím v potravinách a krmivech. V teoretické části jsou také popsány mykotoxiny se společným označením „emerging“, které se v posledním desetiletí staly středem zájmu nejrůznějších výzkumů z důvodu jejich relativně častého výskytu v mnoha komoditách a poměrně mála informací ohledně jejich možných toxických účinků spojené i s absencí metod na jejich stanovení. Praktická část této práce se zaměřuje právě na vytvoření metody pro stanovení tří mykotoxinů ze skupiny „emerging“, a to konkrétně na enniatiny B, enniatin B1 a beauvericin pomocí kapalinové chromatografie s hmotnostní detekcí.

Abstract:

The subject of this thesis is the study of mycotoxins, which are toxic compounds produced by the filamentous fungi of the genus Fusarium. Main focus of its theoretical part is a general introduction of the large group of mycotoxins, classification of the various toxic substances and their most common producers and possible toxic effects. It then briefly characterizes the most significant Fusarium mycotoxins and specifies amounts thereof allowed in food and fodder (if applicable). The theoretical part also describes mycotoxins which share the label „emerging“ and have, in recent years, become a subject of research, due to their relatively common presence in many commodities, lack of information regarding their potential toxic effects, and the lack of a reliable method of determining their presence. The practical part of this work focuses mainly on creating such a method for detecting three mycotoxins from the „emerging“ group – enniatin B, enniatin B1 and beauvericin – using liquid chromatography combined with mass spectrometry.
