Evaluation of cytoprotective activity of phytohormones and their derivates

Abstract:

Friedreichova ataxie je progresivní onemocnění nervového systému. Příčinou je mutace genu kódujícího frataxin, protein, který je nezbytný pro sestavování Fe-S klastrů v některých proteinech dýchacího řetězce. Důsledkem je potom abnormální fungování mitochondrií spojené se sníženou produkcí energie, zvýšeným oxidativním stresem a hromaděním železa. Terapie doposud chybí. V této práci se zabývám možností chránit fibroblasty s mutantním genem pro frataxin proti různým formám exogenně aplikovaného stresu pomocí látek rostlinného původu, především fytohormonů a jejich derivátů. Pro vybrané látky byl také sledován vliv na hladiny redukovaného glutathionu a na aktivitu CSNK1. Získané výsledky poukazují na to, že protektivní účinek aktivních derivátů s inhibicí CSNK1 nesouvisí.

Abstract:

Friedreich´s ataxia is a progressive neurodegenerative disorder. It is caused by a mutation in the gene for frataxin. Frataxin is a protein critical for assembling of Fe-S clusters in several proteins of respiratory chain. Its dysfunction results in an abnormal mitochondrial function associated with a decrease in energy production, increase in oxidative stress and accumulation of iron. Therapy is not available. In this thesis, I explore a possibility of protecting fibroblasts with a mutant frataxin gene against various forms of exogenously applied stress by substances of plant origin, including phytohormones and their derivatives. The effect of selected compounds on the levels of the reduced glutathione and on the activity of CSNK1 was studied as well. The results suggest that the protective effect of active derivatives is not related to inhibition of CSNK1.
