Effect of External Factors on Decarboxylase Activity of Lactobacillus curvatus in vitro and in Milk

Abstract:

Diplomová práce byla zaměřena na prozkoumání dekarboxylázové aktivity bakterií Lactobacillus curvatus. Teoretická část práce obsahuje charakteristiku biogenních aminů, včetně jejich významu, toxických účinků a výskytu v potravinách. Dále se zabývá vnějšími faktory ovlivňujícími produkci biogenních aminů a metodami jejich stanovení. V závěru teoretické části je věnována pozornost charakteristice rodu Lactobacillus a jeho významu v potravinářství. V praktické části byly aplikovány vybrané faktory (pH, obsah NaCl, obsah laktózy a teplota kultivace) ovlivňující růst bakterie Lactobacillus curvatus a její produkci biogenních aminů. Produkce biogenních aminů byla analyzována pomocí vysokoúčinné kapalinové chromatografie s UV/VIS detekcí po derivatizaci danzylchloridem. Testovaná bakterie produkovala pouze tyramin a spermin. Tyramin byl produkován v koncentracích 1,33 až 52,28 mg/l, kdy nejvyšší koncentrace tyraminu bylo dosaženo při iniciačním pH 6,8 v kombinaci s 4,80 % (w/v) laktózy, bez přídavku NaCl a teplotě kultivace při 37 °C. Produkce sperminu byla v průběhu kultivace kolísavá a nejvyšší množství bylo stanoveno při teplotě kultivace 10 °C v podmínkách in vitro bez přídavku NaCl a laktózy.

Abstract:

This thesis was focused on the examination of decarboxylase activity of bacteria Lactobacillus curvatus. Theoretical part of the thesis contains characterization of biogenic ammines, with their significancy, toxic effects and their occurence in food. It also deals with external factors influencing the production of biogenic amines and methods of their determination. In the end of theoretical part, its paid attention to the characteristics of the genus Lactobacillus and its importance in the food industry. In the practical part of the thesis, selected factors were applied (pH, NaCl content, lactose content and cultivation temperature) to affect the growth of bacteria Lactobacillus curvatus and its production of biogenic amines. The production of biogenic amines was analysed by high performance liquid chromatography with UV/VIS detection after derivatization with dansylchloride. The tested bacteria produced only tyramine and spermine. Tyramine was produced in concentrations from 1,33 to 52,28 mg/l, where the highest concentration of tyramine was achieved by initiating pH 6,8 in combination with 4,80 % (w/v) lactose, without the addition of NaCl, and cultured at a temperature 37 °C. Production of spermine was fluctuating during cultivation, and the highest rate was determined at 10 °C, culturing in vitro without the addition of NaCl and lactose.
