Detection of cancer stem cell markers in cell lines derived from pancreatic adenocarcinoma

Abstract:

Adenokarcinom pankreatu (PDAC) je čtvrtou nejčastější příčinou úmrtí v souvislosti s malignitami a pětiletého přežití dosahuje méně než 5 % pacientů. Největší problém představuje především pozdní stanovení diagnózy, kdy už je obvykle primární nádor doprovázený rozsáhlými metastázami, a dále vysoká rezistence tohoto nádoru vůči konvenční chemoterapii a radioterapii. Tyto vlastnosti nádoru, které jsou hlavní příčinou selhání léčby, má pravděpodobně na svědomí malá podskupina nádorových buněk s označením nádorové kmenové buňky (CSCs). K zaměření léčby cíleně na tuto subpopulaci je nejprve třeba jejich fenotypová charakterizace CSCs na základě stanovení specifických povrchových nebo intracelulárních markerů. Diplomová práce je zaměřena na detekci kandidátních markerů CSCs ve vybraných buněčných liniích derivovaných z PDAC - konkrétně se jedná o povrchové antigeny CD44, CD24, epiteliální specifický antigen (ESA) a CD133, a dále o intracelulární protein nestin. Základním metodickým přístupem k řešení byla imunodetekce (nepřímá imunofluoresence, průtoková cytometrie a imunoblotting) a exprese těchto markerů na úrovni mRNA byla detekována pomocí RT-PCR. Na základě výsledků z použitých metod lze říci, že u tří zkoumaných pacientských buněčných linií derivovaných z PDAC byla úspěšně detekována různá míra exprese všech vybraných markerů (CD44, CD24, ESA, CD133 a nestin), které se řadí mezi kandidátní specifické markery CSCs PDAC

Abstract:

Pancreatic adenocarcinoma (PDAC) is the fourth most frequent cause of death related to cancers and five-year survival rate is under 5 %. The main problems concern especially the late diagnosis because the primary tumor is usually accompanied by extensive metastases and the high resistance to conventional chemotherapy and radiotherapy. A small subset of tumor cells termed cancer stem cells (CSCs) is probably responsible for these tumor features, which probably play a key role in the treatment failure. To focus the targeted therapy on this subpopulation of cancer cells, their phenotypic characterization by determination of specific surface or intracellular markers is needed. This diploma thesis is aimed at the detection of putative CSCs markers in selected cell lines derived from PDAC, namely CD44, CD24, epithelial specific antigen (ESA) and CD133 cell surface markers, as well as the intracellular protein nestin were investigated. The main methodological approach was immunodetection (indirect immunofluorescence, flow cytometry, and immunoblotting) and the expression of these markers was also detected at mRNA level by RT-PCR. Based on the achieved results we can conclude that different levels of expression all of these selected putative CSCS markers (CD44, CD24, ESA, CD133 and nestin) in the three examined cell lines derived from PDAC were successfully detected.
