Molecular-genetic analysis of microsatellite variability within the domestic chicken MHC

Abstract:

Hlavní histokompatibilitní komplex (MHC) je oblast v kuřecím genomu, která je nejlépe prozkoumaná a zodpovědná za odolnost vůči chorobám a imunitní reakce u kura. Geny MHC se také podílejí na dalších funkcích, jako jsou produkční vlastnosti a reprodukční schopnosti. Diplomová práce se zabývá studiem variability mikrosatelitových markerů genů pro MHC u kura domácího. Práce se skládá z teoretické a praktické části. Teoretická část obsahuje obecnou charakteristiku užitkovosti a genetické struktury kura domácího. Dále je popsáno využití markerů ve šlechtitelských metodách, charakteristika mikrosatelitů, jejich rozdělení a aplikace. Byla charakterizována MHC oblast kura domácího, jeho molekulární a genetická struktura. Praktická část je věnována ověření a optimalizaci metody pro testování dvou mikrosatelitových lokusů LEI0258 a MCW0371, které se nachází v MHC-B oblasti. Mikrosatelit LEI0258 vykazoval větší polymorfismus než MCW0371. Nejvíc frekventována byla alela 384 bp pro LEI0258. U MCW0371 největší frekvenci vykazovala alela s délkou 205 bp. Obecně lze říct, že optimalizovaná metodika je spolehlivá a že studované markery jsou vhodné pro hodnocení genetických vztahů v populaci a pro studium asociace s jednotlivými znaky.

Abstract:

Major histocompatibility complex (MHC) is the region of the chicken genome that is best studied and responsible for disease resistance and immune responses in chickens. MHC genes are also involved in other functions, such as production traits and reproductive abilities. The diploma thesis deals with the study of the variability of microsatellite markers of MHC genes in the domestic chicken. The thesis consists of theoretical and practical part. The theoretical part contains a general characteristic of the performance and genetic structure of the domestic chicken. Furthermore, the use of markers in breeding methods, characteristics of microsatellites, their distribution and application are described. The MHC region of the domestic chicken, its molecular and genetic structure were characterized. The practical part is devoted to verification and optimization of the method for testing two microsatellite loci LEI0258 and MCW0371, which is located in the MHC-B region. Microsatellite LEI0258 showed greater polymorphism than MCW0371. The 384 bp allele for LEI0258 was the most frequented. In MCW0371, the 205 bp allele had the highest frequency. In general, it can be said that the optimized methodology is reliable and that the studied markers are suitable for the evaluation of genetic relationships in the population and for the study of the association with individual traits.
