Autophagy – cell death or cell survival mechanism?

Abstract:

Autofagie je často řazena mezi 3 základní typy buněčné smrti: apoptóza (typ I) autofagie (typ II) a nekróza (typ III). Je to fyziologický proces zúčastněný v recyklaci organel a buněčného materiálu během pro buňku nepřiznivých podmínek. Jedná se o mechanismus zahrnující tvorbu typických měchýřku a také zapojení lysosomů při samotrávení buněčných organel. V současnosti bylo publikováno mnoho prací ukazujících kontroverzní roli autofagie - na jedné straně jako procesu umožňujícího přežití buněk v nepříznivých podmínkách, na straně druhé procesu, který vede k programované buněčné smrti. Poruchy mechanismu autofagie byly pozorovány u mnoha závažných chorob jako je rakovina, infekční a neurodegenerativní choroby, lysozomální střádavá onemocnení, ale i stárnutí. Pochopení mechanismu autofagie a jeho regulace může napomoci léčbě těchto chorob. Cílem bakalářské práce bude zpracovat literární přehled o autofagii zahrnující přehled základních typů autofagie a stručný popis jejich molekulárních mechanismů. Dále se práce zaměří na rešerši výsledků prací prokazujících význam autofagie pro přežití nebo naopak smrt buněk, vztah autofagie k ostatním typům buněčné smrti (apoptóza, nekroptóza) a na mechanismy a podněty, ktetými může být autofagie vyvolána.

Abstract:

Autophagy is often ranked among the three basic types of cell death: apoptosis (type I), autophagy (type II) and necrosis (type III). It is physiological process involved in recycling of cellular organelles and material during unfavorable conditions for the cell. It is a mechanism involving the formation of vesicles and typical involvement in lysosomes selfdigestion of organelles. Currently many works have been published showing controversial role of autophagy - on the one hand, as a process allowing cell survival in unfavourable conditions, on the other hand process that leads to programmed cell death. Interferences of autophagy mechanism was observed in many serious diseases such as cancer, infectious and neurodegenerative diseases, lysosomal storage diseases, but also aging. Understanding of mechanism and regulation of autophagy may help treat these diseases. The aim of the thesis compile literary overview about autophagy, including an overview of the basic types of autophagy and brief description of their molecular mechanisms. The work focus on the search results of the work demonstrate the importance of autophagy for cell survival and cell death, relationship to other types of cell death (apoptosis, necroptosis) and the mechanisms andstimuli, which can be autophagy induced.
