The effect of combination of nitro-oleic acid and ionizing radiation on survival and signaling of human A549 adenocarcinoma cells

Abstract:

Role a postavení radioterapie v léčbě nádorů se v poslední době vlivem technologického pokroku dostává stále více do popředí. Jedná se o jednu z hlavních metod léčby maligních nádorů. Přestože se neustále zlepšuje terapeutický potenciál metody, její účinnost je stále omezena maximální možnou dávkou záření dodávaného do nádoru, kvůli její toxicitě na okolní zdravou tkáň. Radiorezistence nádorových buněk a její stupňování během radioterapie je jedním z hlavních problémů omezující účinnost léčby. Zvýšení účinnosti radioterapie je možné docílit pomocí radiosenzitizátorů, které zvyšují citlivost nádorových buněk na ionizující záření. Studiu radiosenzitizace/radiorezistence nádorových buněk se proto věnuje velká pozornost. Nitrované mastné kyseliny jsou důležitými regulátory antioxidačních a protizánětlivých procesů v organismu. Studie prováděné na nádorových buňkách poukazují na jejich možný terapeutický účinek, který by mohl napomoci snížit radiorezistenci nádorových buněk. Cílem práce bylo zjistit, zda nitrované mastné kyseliny v kombinaci s γ-zářením vykazují radiosenzitizační účinek na nádorové buňky lidského adenokarcinomu A549. Pro získání experimentálních výsledků byly použity následující metody: MTT test, apoptotický test, test klono-genního přežívání a Western blot. Výsledky naznačují, že NO2OA ovlivňuje reparaci DSB pomocí homologní rekombinace, při studovaných podmínkách se však toto ovlivnění nemanifestovalo v životnosti ozářených buněk, a nevedlo tudíž k radiosensitizaci buněk A549 k ionizujícímu záření. Celkově lze shrnout, že diplomová práce přispěla k lepšímu pochopení vlivu nitrovaných mastných kyselin a γ-záření v biologických procesech u lidské nádorové buněčné linie plicního alveolárního epitelu A549 v podmínkách in vitro.

Abstract:

The role of radiotherapy becomes more prominent due to technological advancement in cancer treatment. It is one of the main treatment methods for malignant tumours. Although the therapeutic potential of the method is constantly improving, its effectiveness is still limited by the maximal dose of radiation delivered to the tumour, due to its toxicity to the surrounding healthy tissue. The radioresistance of cancer cells and escalation is one of the main issues limiting the effec-tiveness of radiotherapy during cancer treatment. The enhancement of radiotherapy's effectiveness is possible to achieve utilizing radiosensitizers which enhance the sensibility of cancer cells to ionizing radiation. Major attention is paid to the field of radiosensitization/ radiore-sistence of cancer cells. Nitro-fatty acids are important regulators of antioxidant and anti-inflammatory processes in an organism. The experiments conducted on cancer cells indicate their possible therapeutic effect which could be helpful to decrease cancer cells’ radioresistance. The thesis aims to find out whether the nitro-fatty acids in combination with γ-radiation show a radiosensitizing effect on cancer cells of human adenocarcinoma A549. These methods were used for gaining experimental results: MTT test, apoptotic test, clonogenic survival test and Western blot. The results indicate that NO2OA affects DSB repair by homologous recombination, but under the specified conditions, this effect did not manifest itself in the viability of the irradiated cells and thus did not lead to radiosensitizing effect on A549 cells to ionizing radiation. Overall, the thesis contributed to a better understanding of the effect of nitro-fatty acids and γ-radiation in biological processes in the human lung alveolar epithelial tumour cell line A549 in vitro.
