“Cell free” expression systems and their adoption for production of parasitic worms recombinant proteins

Abstract:

Cell-free (CF) proteinová exprese má své počátky v 60. letech minulého století. Od té doby se metoda významně zdokonalila v mnoha směrech. Byly například vylepšeny buněčné extrakty, na kterých celá metoda stojí, a také byl optimalizován systém regenerace energie, nezbytný pro správný průběh reakce. Obé pomohlo dosáhnout o několik řádů vyšších proteinových výtěžků, než v počátcích, což dalo vzniknout komerčně dostupným kitům obsahujícím vše potřebné pro reakci. V současné době jsou k dostání mnoho variant kitů od mnoha společností. Jednotlivé kity se liší hlavně v použitém buněčném extraktu a množstvím dodávaných reagencií. Za účelem experimentálního ověření využitelnosti CF kitu pro přípravu rekombinantních proteinů helmintů byly vybrány tři funkční proteiny, dvou modelových organismů - katepsin L Eudiplozoon nipponicum (Monogenea) a katepsiny F a B Trichinella spiralis (Nematoda). Katepsiny jsou cysteinové peptidázy, významné enzymy, využívané helminty k degradaci či destrukci hostitelských tkání v průběhu penetrace či migrace a jsou zapojené také do procesu trávení a podílí se i na dalších významných biologických pochodech, nezbytných pro život parazitických helmintů.

Abstract:

Since 1960, the beginning of cell-free (CF) protein expression approaches, the method has been significantly improved in many ways. For example the cell extracts, has been optimized as well as an energy renewal system, which is very important for the proper function of CF expression. Both improvements helped to achieve considerably higher protein yield. Subsequently this initiated the production of commercially available kits, which contained all the necessary components for the reaction. Currently, many kits of number of companies are available and differ mainly in type of used cell extract and amount of supplied reagents (number of reactions). Expressway CF E. coli kit was used for production of parasitic worms’ recombinant proteins. Three functional proteins from two model organisms were chosen – cathepsin L of Eudiplozoon nipponicum (Monogenea) and cathepsins F and B of Trichinella spiralis (Nematoda). Cathepsins are cysteine peptidases, the essential enzymes used by helminths for tissue degradation during penetration or migration within the host and they are also related to digestion and other important biological processes.
