Use of capillary electrophoresis with laser induced fluorescence detection in analysis of biomarkers

Abstract:

Obecně se tato disertační práce zabývá vývojem metod kapilární elektroforézy (CE) pro analýzu potenciálních diagnostických biomarkerů v neinvazivních biologických tekutinách. Neinvazivní biologické tekutiny, jako je například kondenzát vydechovaného vzduchu (EBC), obsahují obvykle nízké koncentrace biomarkerů a je proto vhodné použití vysoce citlivých analytických metod. Tato disertační práce popisuje tři různé CE metody. V první části je popsána konstrukce nové kompaktní, vysoce citlivé CE s laserově indukovanou fluorescencí (LIF). Následně byl přístroj CE-LIF použit k vývoji a optimalizaci nové citlivé metody pro stanovení malondialdehydu (MDA) v EBC a dalších biologických tekutinách s velmi nízkým detekčním limitem (LOD ~ 1,1 nM). Druhá část byla zaměřena na optimalizaci složení základního elektrolytu pro simultánní separaci a detekci markerů nitrosativního a oxidačního stresu pomocí duální C4D-LIF detekce. Poslední část byla věnována vývoji alternativní metody pro diagnostiku refluxní choroby jícnu (GERD). V této části byl srovnáván iontový profil a pH vzorku EBC u pacientů s GERD a zdravými kontrolami. Zjistili jsme, že vybrané ionty, zejména sodný kationt, butyrát a propionát, byly zvýšeny ve vzorcích EBC pacientů trpících GERD.

Abstract:

In general, this dissertation thesis deals with the development of capillary electrophoresis methods for the analysis of potential diagnostic biomarkers in non-invasive biological fluids. Non-invasive biological fluids, such as exhaled breath condensate (EBC), usually contain low concentrations of biomarkers; therefore, the use of a highly sensitive analytical method is necessary. This dissertation thesis describes different approaches for such method development. In the first part the construction of a new compact, highly sensitive capillary electrophoresis (CE) instrument with laser induced fluorescence (LIF) is described. The CE-LIF instrument is used for the optimization of a new sensitive method for the determination of malondialdehyde (MDA) in EBC and other biological fluids with limits of detection (LODs) as low as 1.1 nM. The second part of the thesis is focused on optimizing the background electrolyte for simultaneous separation and detection of nitrosative and oxidative stress markers using dual C4D-LIF detection mode. The last part is dedicated to the development of an alternative diagnostic method for diagnosing gastroesophageal reflux disease (GERD). The ionic profile and pH of the EBC samples were compared in patients with GERD and healthy controls. Selected ions, mainly sodium, butyrate, and propionate, were elevated in EBC samples from patients suffering from GERD.
