Changes in leaf optical properties in selected evergreen plants in response to UV-B radiation

Abstract:

Tato diplomová práce se zabývá vlivem UV-B záření a v menším rozsahu vlivem UV-A záření na rostliny, s důrazem na aktivaci biochemických a biofyzikálních ochranných mechanismů vůči negativním účinkům UV záření. Práce v tomto ohledu představuje úvod do problematiky skupiny látek nazývané flavonoidy, jejichž vlastnosti jsou zkoumány z hlediska možnosti odstínění UV záření a zachycování reaktivních forem kyslíku (ROS). Experiment vyhodnocuje některé vlivy UV-B záření na vybrané sukulentní rostliny: Sedum album ’Athoum’, Sedum sexangulare, and dva neurčené kultivary Sempervivum sp. Zkoumanými parametry jsou: obsah antokyanů v listech ovlivněných UV-B zářením a reakce fluorometrických parametrů v rámci primárních fotosyntetických procesů. Obsah antokyanů byl měřen pomocí změn v reflektanci listů a pomocí spektrofotometrické metody. Fluorometrické parametry primárních fotosyntetických procesů byly měřeny CCD kamerou zachycující fluorescenční kinetiku.

Abstract:

This thesis study effects of UV-B and to a lesser degree UV-A radiation on plants with emphasis on the activation of biochemical and biophysical mechanisms of protection against harmful effects of UV radiation. In this context, the thesis provides a basic review of literature about flavonoid compound group. The properties of these biomolecules are often considered to be useful in UV screening and ROS (reactive oxygen species) scavenging. The experiment evaluates some effects of artificial supplementary UV-B radiation on selected succulent plants: Sedum album ’Athoum’,Sedum sexangulare, and two undetermined cultivars of Sempervivum sp. The examined parameters were: anthocyanin content in a UV-B treated leaf, and the response of chlorophyll fluorescence parameters within the primary photosynthetic processes. The anthocyanin content was measured by changes in reflectance of leaves and by spectrophotometric analysis. The fluorometric parameters of primary photosynthetic processes were measured by a chlorophyll fluorescence kinetics CCD camera.
