Serotonergic psychedelics - LSD, psilocybin and DMT, and their therapeutic use.

Abstract:

Práce se věnuje 3 vybraným serotonergním psychedelikům – LSD, psilocybinu a DMT, se zaměřením na jejich terapeutické využití. Jde o dynamicky se rozvíjející doménu vědeckého bádání zejména v posledních letech. Výzkumná část má podobu přehledové studie, v níž je zpracován ucelený a systematický přehled prokazatelného terapeutického využití těchto tří psychedelik. V teoretické části je poskytnut dostatek informací pro pochopení základních souvislostí tohoto komplexního tématu. Studie zařazené do přehledu byly získány vyhledáváním ve 4 akademických odborných databázích a také manuálně. Získané studie byly zkoumány především z hlediska dvou aspektů – metodologické úrovně a terapeutického využití. Pro DMT nebyly nalezeny žádné relevantní studie. Studií s psilocybinem byl dvojnásobný počet oproti těm s LSD a vykazovaly celkově vyšší úroveň metodologického standardu. Prokazatelné terapeutické využití bylo dokumentováno u LSD i psilocybinu. U LSD byl nalezen terapeutický efekt například v oblasti úzkosti při život ohrožujícím onemocnění, osobnostních charakteristik, pozitivních životních změn, postojů a nálady, sugestibility či mentální imaginace. U psilocybinu byl terapeutický efekt nalezen kupříkladu v oblasti úzkosti a deprese při život ohrožujícím onemocnění nebo při farmakorezistentní depresi, obsedantně kompulzivní symptomatiky, závislostních tendencí u alkoholu a tabáku, osobnostních charakteristik, pozitivních životních změn, postojů a nálady či emocionální empatie.

Abstract:

The thesis deals with 3 selected serotonergic psychedelics – LSD, psilocybin and DMT, focusing on their therapeutic use. It is a dynamically developing field of scientific research, especially in recent years. The research part is in the form of a review study in which a comprehensive and systematic review of the proven therapeutic use of these three psychedelics is elaborated. The theoretical part provides sufficient information to understand the basic context of this complex topic. The studies included in the review were obtained by searching in 4 academic professional databases and also manually. The obtained studies were examined mainly from the point of view of two aspects – level of methodology and therapeutic use. No relevant studies were found for DMT. Number of psilocybin studies was twice as high as those with LSD and psilocybin studies possessed generally higher methodological quality. Evidence of therapeutic use has been documented in both LSD and psilocybin. LSD has been found to have a therapeutic effect in the domains of anxiety in life-threatening diseases, personality traits, positive life changes, attitudes and mood, suggestibility, mental imagination and others. In psilocybin, the therapeutic effect was found in domains of anxiety and depression in life-threatening disease or in pharmacoresistant depression, obsessive-compulsive symptomatology, alcohol and tobacco addiction tendencies, personality traits, positive life changes, attitudes and mood, emotional empathy, and other areas.
