Telomeric-sequence associated proteins a their function

Abstract:

Telomery jsou DNA-proteinové komplexy na koncích lineárních eukaryotických chromozomů. Skládají se z telomerické sekvence a proteinů asociovaných s touto sekvencí. Funkce telomer spočívá v ochraně konců chromozomů a zamezení replikativnímu zkracování konců chromozomů. Zkracování telomer zabraňuje enzym telomeráza, který má schopnost prodlužovat telomery. Ochranu telomer má u savců na starosti proteinový komplex zvaný shelterin. U rostlin je známé množství kandidátních proteinů, ale přesné složení komplexu chránícího telomery ještě nebylo zcela rozřešeno. Nejlépe je proteinové složení telomer prozkoumáno u Arabidopsis thaliana. Kromě shelterinu se na telomery váže další proteinový komplex – CST komplex. Ten má u rostlin funkci ochrany telomer, u savců je výraznější jeho role při replikaci. Kromě telomerických funkcí byla u mnoha telomerických proteinů objevena i funkce mimo telomery. Mnoho z nich funguje například jako regulátory exprese.

Abstract:

Telomeres are nucleoprotein complexes at the ends of linear eukaryotic chromosomes. They consist of a telomeric DNA sequence and proteins associated with this sequence. The role of telomeres is to protect the chromosome ends from damage and replicative shortening. Shortening of telomeres is prevented by the action of telomerase, an enzyme that has the ability to prolong telomeres. Protection of telomeres from damage is mediated by the shelterin complex in mammals. A number of candidate proteins are known in plants, however the exact composition of the telomere protection complex has not yet been resolved. The protein composition of telomeres has been best examined in Arabidopsis thaliana. Another protein complex that binds the telomeres apart from shelterin is the CST complex. The CST complex has an end protective function in plants, but in mammals, it has a more prominent role in replication. Apart from telomeric functions, many telomeric proteins also have non-telomeric functions. For example, many telomeric proteins have been shown to regulate transcription.
