Long-term mild hyperthermia in cancer treatment

Abstract:

Diplomová práce je zaměřena na hypertermickou léčbu, především mírně zvýšenými teplotami a její využití v léčbě rakoviny. Práce se zabývá přístupy, jak dosáhnout zvýšení teplot a následné léčby u pacientů, mechanismy působení hypertermie a možnostmi využití hypertermie s dalšími léčebnými postupy. Dále se zabývá problematickými aspekty aplikace vysokých nefyziologických teplot a výhodami dlouhodobé mírné hypertermie. Tato páce odůvodňuje využití hypertermie při léčbě onkologických onemocnění a zdůrazňuje její výhody. Experimentální část práce je zaměřena na účinek dlouhodobé mírné hypertermie na řadu nádorových a nenádorových buněčných linií. To zahrnuje hodnocení buněčné proliferace, toxického efektu na buňky, stresové signalizace, odpovědi na teplotní poškození (tzv. "heat shock protein response"), odpovědi na poškození DNA, detekci replikujících se buněk a analýzu buněčného cyklu. Výsledky ukazují cytostatický a také cytotoxický účinek dlouhodobé mírné hypertermie v řadě nádorových buněčných liniích, zatímco nenádorové buňky jsou k tomuto typu léčby více rezistentní. Kromě výše zmíněného ukazují získaná data schopnost dlouhodobé mírné hypertermie potenciovat efekt genotoxické látky olaparibu.

Abstract:

The diploma thesis is focused on hyperthermic treatment, especially with mild temperatures, and its use in cancer therapy. The thesis deals with approaches, how to reach hyperthermia treatment in patients, mechanisms of action, and possibilities of hyperthermia usage as adjuvant therapy with other treatment modalities. We discuss the problematic aspects of applying high non-physiological temperatures and the advantages of long-term mild hyperthermia. The thesis underlines the rationale for the utilization of hyperthermia in cancer therapy. The experimental part of the thesis reports the effect of long-term mild hyperthermia on various cancerous and non-cancerous cell lines. These comprise evaluating cell proliferation, a cytotoxic effect on cells, stress signalling, heat shock protein response, DNA damage response, detection of replicating cells, and cell cycle analysis. The results show a cytostatic and also cytotoxic effect of long-term mild hyperthermia for numerous cancerous cell lines, whereas non-cancerous cells are more resistant to this treatment. In addition to this, acquired data demonstrate the capability of long-term mild hyperthermia to potentiate the effect of the genotoxic agent olaparib.
