Preparation of lectin PSL2 from Pisum sativum

Abstract:

This thesis is focused on the production optimization and purification of the hypothetical PSL2 lectin and subsequent structural - functional studies. Lectins are proteins reversibly binding carbohydrates. Among other things, they are characterized by the fact that they are able to agglutinate erythrocytes. They are present across animal, plant, fungal and bacterial organisms, including protozoa. Thanks to their major occurence and variability of functions and features, they have wide range of applications. To obtain this PSL2 protein, various strains of E. coli cells were transformed by two kinds of constructs. The first construct was previously prepared in our laboratory. It contained the gene for thioredoxin fusion protein, which is not only conducive to the protein solubility of the target PSL2 protein, but also works as a tag by which the recombinant protein can been purified. A second construct was designed and prepared in this Master´s thesis. It contained the gene for the highly soluble NusA fusion protein, which was used to advance the production of a target PSL2 protein in his soluble form. Two oligohistidine tags on the N-terminal sequence of the psl2 gene were used to purify the PSL2 protein with NusA fusion protein. Conditions for production of the recombinant PSL2 protein were optimized by the E. coli cells modified by the two selected constructs. Further, the optimization of the purification using the method FPLC with metaloaffinity chromatography was performed using Ni-NTA column. The protein obtained by purification was subjected to structural - functional studies. Also, two constructs for production of the PSL2 protein in eukaryotic expression cells L. tarentolae were proposed and prepared.

Abstract:

Tato diplomová práce je zaměřena na optimalizaci produkce a purifikace hypotetického lektinu PSL2 a na jeho následné strukturně - funkční studie. Lektiny jsou proteiny reverzibilně vázající sacharidy. Mimo jiné se vyznačují i tím, že jsou schopny aglutinovat erytrocyty. Vyskytují se napříč živočišnou, rostlinnou, houbovou a bakteriální říší, včetně prvoků. Díky rozsáhlému výskytu a variabilitě funkcí a vlastností mají lektiny široké využití. Na zisk zmíněného proteinu PSL2 byly využity různé kmeny bakteriálních buněk E. coli, které byly transformovány dvěma druhy DNA konstruktů. První konstrukt byl již dříve připraven v naší laboratoři. Obsahoval gen pro fúzní protein tioredoxín, který sloužil nejen jako protein napomáhající rozpustnosti cílového proteinu PSL2, ale zároveň i jako kotva, pomocí které byl tento rekombinantní protein purifikován. Druhý konstrukt byl navržen a připraven v rámci této práce. Obsahoval gen pro vysoce rozpustný NusA fúzní protein, který sloužil na posun produkce cílového proteinu PSL2 do rozpustné formy. Na purifikaci proteinu PSL2 s Nusa fúzním proteinem byly použity dvě oligohistidínové kotvy, které byly připojeny na N-konec sekvence pro gen psl2. Pomocí takto modifikovaných buněk E. coli prostřednictvím dvou vybraných konstruktů byly optimalizovány podmínky produkce rekombinantního proteinu PSL2. Dále byla provedena optimalizace purifikace proteinu PSL2 pomocí metody FPLC s využitím metaloafinitní chromatografie na Ni-NTA koloně. Protein získaný purifikací byl podroben strukturně - funkčním studiím. Na produkci proteinu PSL2 v eukaryotickým expresním organismu L. tarentolae byly také navrženy a připraveny dva konstrukty.
