Abstract:

Předmětem této disertační práce je studium složení močových kamenů, u kterých znalost prostorové distribuce matricových a stopových prvků může pomoci vysvětlit příčinu jejich vzniku a následného růstu. Pro účely mapování prostorového složení bylo využito několika metod a to především laserové ablace s indukčně vázaným plazmatem s hmotnostní nebo optickou emisní spektrometrií (LA-ICP-MS/OES), spektrometrie laserem buzeného mikroplazmatu (LIBS) a rentgenové mikrofluorescence buzené synchrotronovým zářením (SR-µXRF). Byla studována vhodnost použití laserové ablace v režimu liniového skenu a rastru bodů a bylo zjištěno, že liniový sken je vhodný u větších vzorků a naopak rastr bodů pro zachycení detailu menších oblastí. Stěžejním bodem prostorové analýzy močových kamenů laserovými technikami je kvantifikace získaných distribučních prvkových 2D map. Z důvodu nedostatku standardních referenčních materiálů byla pro tyto vzorky využita metoda kalibrace za pomoci reálných vzorků močových kamenů, tzv. standardy s přizpůsobenou matricí. Pro ověření správnosti výsledků sloužila elektronová mikrosonda. Vylisované tablety ze vzorků močových kamenů také sloužily ke srovnávací studii různých laserových technik. Metoda LIBS byla také použita pro studium prostorové distribuce matričních prvků u vzorků dolních čelistí chameleona jemenského. Dochází zde ke vzniku akrodontní ankylózy, což je pevné spojení mezi zubem a čelistí.

Abstract:

The aim of this work presented in this thesis is the study of urinary stones composition. The knowledge of their spatial distribution of the matrix and trace elements can help to explain the cause of their formation and subsequent growth. For the purpose of mapping spatial composition were used some methods, namely laser ablation inductively coupled plasma mass spectrometry and optical emission spectrometry (LA-ICP-MS / OES), laser induced breakdown spectrometry (LIBS) and X-ray microfluorescence excited by synchrotron radiation (SR μXRF). In this work the suitability of laser ablation in the scheme of line scan and grid points were studied. It was found that the line scan is suitable for use with larger samples and vice versa grid points to capture the details of smaller areas. The main task of the spatial analysis of urinary stones by laser ablation techniques is the quantification of obtained elemental distributive 2D maps. Due to the lack of standard reference materials for these samples calibration method, so called matrix-matched standards with the help of real samples of urinary stones were used. To verify the accuracy of the results electron microprobe was used. The compressed pellets from samples of urinary stones also served for a comparative study of various techniques based on laser ablation. LIBS method was also used to study the spatial distribution of matrix elements in samples of the lower jaw of veiled chameleon. There has been a formation of acrodont ankylosis, which is a fixed connection between the tooth and jaw.
