Impact of histones acetylation on the stability of plant telomeres

Abstract:

In eukaryotic cells, DNA is organized into a complex structure called chromatin. Chromatin undergoes epigenetic regulation which is accomplished mainly by modifications of DNA and histone. The acetylation of histones impacts significantly on chromatin structure and gene expression. In the theoretical part of the diploma thesis „Impact of histones acetylation on the stability of plant telomeres“ knowledge about epigenetic mechanisms, especially about histone modifications was summarized. The second part dealt with structural regions located at chromosome ends – telomeres, their structure and epigenetic regulation. The experimental part of the diploma thesis was focused on the study of Arabidopsis thaliana mutant plants with T-DNA insertion in the gene encoding HAC1, HAC5 or HAC12 histone acetyltransferase and plants with mutations in the HDA6 histone deacetylase. It was found that disruption of HAC genes function did not affect the telomeres length; the activity of telomerase, an enzyme responsible for maintaining a stable telomer length, was even increased in the hac12-1 line, in correlation with increased level of the transcript of the AtTERT gene coding for the telomerase protein subunit. In both hda mutant lines telomeres were significantly shortened, but this phenotype was not correlated with a change of the telomerase activity or the relative level of transcripts of the AtTERT gene. In both types of mutants, certain changes of the relative gene transcripts of genes encoding proteins functionally associated with telomere or telomerase were detected; correlation of these changes with the observed telomeric phenotype was discussed. Analysis of telomeric histones modifications in the hda6 lines showed a decrease of the relative level of heterochromatin-specific H3K9me2 modification and an increase of the relative level of H3K27me3, a modification referring to developmentally silenced genes; both modifications were previously identified on plant telomeres. We concluded, that the loss of function of proteins responsible for the stable histone acetylation influenced the maintenance of telomeres in the Arabidopsis thaliana model plant.

Abstract:

V eukaryotických buňkách je DNA organizována do komplexní struktury zvané chromatin. Chromatin podléhá epigenetické regulaci, která je realizována modifikacemi histonů a bází DNA. Významný vliv na strukturu chromatinu a expresi genů má acetylace histonů. Diplomová práce "Vliv acetylace histonů na stabilitu rostlinných telomer" se v teoretické části věnuje shrnutí poznatků o epigenetických mechanismech se zaměřením na modifikace histonů. Další část se zabývá problematikou konců chromosomů – telomerami, jejich strukturou a epigenetickou regulací. Experimentální část diplomové práce je věnována studiu mutantných rostlin Arabidopsis thaliana s inzercí T-DNA v genech kódujících histon acetyltransferázy HAC1, HAC5, nebo HAC12 a s mutací v genu kódujícím histon deacetylázu HDA6. Bylo zjištěno, že porucha funkce genů HAC nemá vliv na délku telomer; aktivita telomerázy, enzymu zodpovědného za udržovaní stabilní délky telomer, byla dokonce zvýšená u linie hac12-1, což korelovalo se zvýšením hladiny transkriptů genu AtTERT, ktorý kóduje proteinovou podjednotku telomerázy. U mutantnů hda byly u obou analyzovaných linií detekovány zkrácené teloméry, nicméně nebyla pozorována významná změna aktivity telomerázy ani relativní hladiny transkriptů genu AtTERT. V případě obou typů mutantů byly detekovány změny relativní hladiny transkriptů některých genů kódujících proteiny funkčně spojené s telomerami či telomerázou; korelace těchto změn s pozorovaným telomérovým fenotypem je diskutována. Analýza modifikací telomerových histonů v liniích hda6 ukázala pokles relativní hladiny modifikace H3K9me2, která je typická pro heterochromatinové úseky, a zvýšení relativní hladiny modifikace H3K27me3, což je modifikace označující vývojově umlčené geny; obě tyto modifikace byly v dřívějších experimentech identifikovány na rostlinných telomerách. Z těchto výsledků vyplývá, že ztráta funkce proteinů zodpovědných za stabilní acetylaci histonů má vliv na stabilitu telomer v modelové rostlině Arabidopsis thaliana.
