Glycoprotein analysis using modified nanoparticles

Abstract:

Diabetes mellitus patří mezi nejrozšířenější a velmi závažná onemocnění. Průvodním jevem této nemoci je glykace proteinů krevní plasmy, které se stanovují pro zjištění kompenzace diabetu. Nejdůležitější z nich je glykovaný hemoglobin (GHb), který je stabilním a specifickým analytem. Jako HbA1c se označuje frakce hemoglobinu, kde je na N terminálním konci navázaná glukosa. Využívá se pro určení dlouhodobé kompenzace diabetu za poslední 3 měsíce. Pro stanovení GHb i jiných glykovaných proteinů je osvědčenou metodou boronátová afinitní chromatografie. Při analýze se využívá afinity derivátů kyseliny borité k cis diolovému uskupení stanovovaných molekul. Tato diplomová práce se zabývá stanovením glykovaných proteinů, zejména glykovaného hemoglobinu, s využitím nanočástic modifikovaných konjugáty s kyselinou boritou. Nejprve byla práce zaměřena na glykační a konjugační postupy, dále na optimalizace stanovení boronátovou afinitní chromatografií, syntézu nanočástic a poté i na jejich modifikaci. Nakonec byla práce soustředěna na vývoj metody pro stanovení obsahu GHb s použitím nanočástic modifikovaných konjugátem kyseliny m-aminofenylborité.

Abstract:

Diabetes Mellitus is one of the most common diseases of people over 50, with a serious impact on the health state of the patients. Higher level of blood glucose concentration leads to nonspecific glycation of plasma proteins and proteins of body organs. Analysis of glycated plasma proteins is used in long-term monitoring of the disease progress, however does not have significant impact on health state. On the other hand, glycation of proteins in body organs structure leads to the serious health problems, even to organ failure, if the disease is not appropriately compensated. Glycated hemoglobin (GHb) is probably the most significant analyte connected with the long term monitoring, fraction HbA1c (hemoglobin glycated on both N-terminal valines) is the GHb fraction recommended by International Committee to be analysed in this case. The diploma thesis was focused on analysis of glycated proteins, especially glycated hemoglobin, based on nanoparticles modified with protein-boronic acid derivative conjugates. First, the glycation of proteins was optimized and studied by various methods including affinity chromatography, mass spectrometry and spectrophotometric methods. Next, synthesis of nanoparticles and their coverage with proteins was performed. Finally, the analytical method based on nanoparticle agglutination was developed.
