Role rostlinných hormonů v lokální a systemické kontrole kořenové architektury při různých růstových podminkách

Abstract:

Pro růst a vývoj rostlin je významné udržování kořenové architektury. Na změnách kořenové architektury se významně podílí fytohormony a jejich interakce. Předchozí studie ukázaly, že cytokininy i auxiny hrají v tomto procesu roli a jejich interakce je zvláště důležitá. Úspěšnou metodou studia vlastností a růstu kořenů je systém rozdělení hlavního kořene, který umožňuje napodobit přirozené heterogenní podmínky v půdě. Možnost využití této metodiky při studiu zasolení byla ověřena a následně byla testována u transgenních rostlin se sníženou (CKX) a zvýšenou (IPT) hladinou cytokininů. Rostliny se zvýšenou a sníženou hladinou cytokininů byly kultivovány tak, že polovina kořenového systému byla v podmínkách zvýšené salinity a druhá polovina v kontrolních podmínkách. Schopnost rostlin modifikovat kořenovou architekturu v reakci na salinitu byla významně snížena u linie IPT akumulující cytokininy. Naopak rostliny linie CKX se sníženou hladinou cytokininů byly stále schopny podporovat růst kořenů v médiu bez přídavku soli. Exogenní aplikace IAA k IPT rostlinám za účelem zvýšení poměru auxin/cytokinin nestačilo k úplnému obnovení schopnosti rostlin podporovat růst kořenů. Výsledky ukazují, že udržení správné hormonální homeostázy je nutné pro adekvátní odpovědi kořenové architektury v heterogenním prostředí. Vysvětlení molekulárního mechanismu těchto jevů však vyžaduje další výzkum.

Abstract:

Maintaining root architecture is important for plant survival. Plants have adapted to alter their root architecture via many hormonal interactions and pathways. Previous studies have shown that cytokinins and auxins play a role in that process, and their interaction is particularly relevant. For better studying the role that these phytohormones play in shaping root architecture in response to heterogeneous environments, the use of a split-root system was chosen, since it could mimic the heterogeneous conditions that plant faces in nature. The setup for the split-root system for salinity treatment was validated, and cytokinin deficient (CKX) and overaccumulating (IPT) transgenic lines were then used to check if cytokinin imbalance affects the plant's ability to properly modify its root architecture in response to salinity. 7 IPT and CKX plants were grown with half of the root system placed in salinity-treated media and the other half in non-treated media. Plant’s ability to modify root architecture in response to salinity was significantly decreased in cytokinin-accumulating IPT lines. On the contrary, cytokinin-deficient CKX plants were still able to favor root growth into non-saline patches of media. The addition of exogenous IAA to IPT plants, in order to raise the auxin/cytokinin ratio was not enough to completely restore the ability of plants to favor growth on non-saline patches of media. However, these results were not enough to adequately evaluate this effect and further research will be required. We can conclude that the maintenance of proper hormonal homeostasis is necessary for adequate adaptation of the root architecture to an heterogeneous environment.
