Possibilities of detection of pathogens of the species Tilletia in seeds of Common wheat and Spelt

Abstract:

Předmětem této diplomové práce je detekce patogenů rodu Tilletia, identifikace rozdílů mezi druhy T. caries a T. controversa a možnosti kvantifikace množství patogena ve vzorcích osiva pomocí molekulárních metod. V literární části jsem se jednotlivých kapitolách zaměřila na popis pšenice seté (Triticum aestivum), pšenice špaldy (Triticum spelta), a především na patogeny rodu Tilletia a možnosti jejich detekce. V práci jsou dále uvedeny způsoby ochrany pomocí mořidel, ale také možnosti ochrany v systému ekologického zemědělství. V jednotlivých kapitolách je také kladen zvláštní důraz na kontrolu osiva. V praktické části byla provedena PCR reakce za použití RPB2 primerů, která potvrdila, že jsou tyto primery jsou vyhovující pro detekci a následnou identifikaci pomocí RFLP analýzy, podle které byla zjištěna přítomnost patogena druhu T. caries. Součástí experimentální části byla také kvantitativní PCR s primery TILLGS2A. Reakce s primery TILGS2A měla vyšší citlivost než reakce s primery RPB2. Také byla zkoušena relativní kvantifikace, byly porovnávány specifické signály s hodnotami reakce vzhledem k vnitřnímu kalibrátoru. Výsledkem bylo potvrzení infekce u některých testovaných vzorků a orientační vyhodnocení množství patogena.

Abstract:

The subject of this diploma thesis is the detection of pathogens of the species Tilletia, the identification of differences between the species T. caries and T. controversa and the possibility of quantifying the amount of pathogens in seed samples using molecular methods. In the literary part, I have in the individual chapters the possibilities for the description of Common wheat (Triticum aestivum), Spelt (Triticum spelta) and especially on pathogens of the species Tilletia and the possibilities of their detection. The work also presents the methods of protection using mordants, but also the possibilities of protection in the system of organic farming. Special emphasis is also placed on seed control in the individual chapters. In the practical part, PCR reaction was performed using RPB2 primers, which confirmed that these primers are suitable for detection and subsequent identification by RFLP analysis, according to which the presence of a pathogen of the species T. caries was detected. The experimental part also included quantitative PCR with primers TILLGS2A. The reaction with primers TILGS2A was more sensitive than the reaction with primers RPB2.. Relative quantification was also tested, specific signals were compared with response values with respect to the internal calibrator. was the confirmation of infection in some tested samples and an indicative evaluation of the amount of pathogen according to.
