Vliv katepsinu D na protinádorové účinky látky TNF-related apoptosis-inducing ligand (TRAIL) v lidských buňkách

The effect of Cathepsin D on anticancer activity of the TNF-related apoptosis-inducing ligand (TRAIL) in human cells

Abstract:

Lysozomální proteáza katepsin D je exprimována téměř ve všech tkáních, účastní se regulace celé řady buněčných dějů a má důležitou roli mimo jiné ve vývoji a přestavbě prsní tkáně. Deregulace katepsinu D je významným faktorem v procesu kancerogeneze. Jeho zvýšená exprese a sekrece byla detekována nejen u prsních karcinomů, ale i u řady dalších typů nádorů. Role katepsinu D v nádorech je dvojí: katepsin D podporuje proliferaci, invazivitu a migraci nádorových buněk pokud je sekretován do extracelulárního prostředí, ale zároveň je mediátorem apoptotické buněčné smrti indukované chemoterapeutiky a jinými stresovými stimuly. Molekulární mechanismus zapojení katepsinu D do apoptotických signálních drah v nádorových buňkách není zcela objasněn. Kromě štěpení specifických substrátů některé studie připouštějí úlohu meziproteinových interakcí v regulaci apoptózy zprostředkované katepsinem D. TRAIL je cytokin, který vazbou na specifický membránový receptor aktivuje apoptotickou signální dráhu v buňce. TRAIL je nadějným chemoterapeutikem zejména pro svou specifickou cytotoxicitu vůči nádorovým buňkám. Cílem této práce bylo charakterizovat funkci katepsinu D v protinádorových účincích cytokinu TRAIL se zaměřením na buňky prsního karcinomu. Pro studium role enzymatické aktivity katepsinu D jsme jako modelový systém vytvořili varianty buněčné linie MDA-MB-231 odvozené z prsního karcinomu, které exprimují exogenní katepsin D nebo jeho enzymaticky inaktivní variantu. Zjistili jsme, že pro-apoptotický účinek katepsinu D je zprostředkován jeho enzymatickou aktivitou. Zkoumali jsme význam zapojení kaspáz a proteinů rodiny Bcl-2 v této apoptotické signalizaci. Posílení aktivace iniciační kaspázy 8, která přímo aktivuje efektorovou kaspázu 3 se jeví jako molekulární mechanismus pro-apoptotického účinku katepsinu D, zatímco destabilizace mitochondrií prostřednictvím aktivovaného proteinu Bid až následně amplifikuje apoptotický signál. Jak TRAILem indukovaná apoptóza tak pro-apoptotický účinek katepsinu D je regulován acidifikací váčků endolysozomálního systému. Také jsme se zabývali možnostmi použití a srovnáním účinnosti inhibitorů katepsinu D v nádorových buňkách. Výsledky této práce přispívají k poznání funkce lysozomální proteázy katepsinu D v regulaci vnější apoptózy indukované TRAILem a identifikují enzymatickou aktivitu katepsinu D jako faktor, který ovlivňuje cytotoxicitu TRAILu vůči prsním nádorovým buňkám.

Abstract:

Cathepsin D, an ubiquitously expressed lysosomal aspartic protease, is involved in a number of biological processes including development and remodelling of mammary gland. Its deregulation is associated with a number of pathological states such as cancer. Cathepsin D is up-regulated in human breast carcinoma and many other tumour types. The role of cathepsin D in tumour cells is dual: secreted cathepsin D stimulates cell proliferation and facilitates cancer cell migration and invasion, while intracellular cathepsin D has been repeatedly reported to act as mediator of apoptosis induced by various chemotherapeutics and other stimuli. The molecular mechanism responsible for the regulation of the chemotherapy-induced cell death by cathepsin D has not been fully elucidated. There is still controversy over the role of enzymatic/proteolytic vs. protein-protein interaction activities. TRAIL is a cytokine, capable of binding to specific membrane-bound receptors to induce apoptotic cell death. It induces apoptosis preferentially in transformed cells without significant toxicity toward normal tissues. The aim of this study was to clarify the role of the cathepsin D in the anticancer activity of TRAIL in breast carcinoma cells. For this purpose, MDA-MB-231 breast carcinoma cells with an increased level of wild type cathepsin D or its mutant enzymatically inactive counterpart were prepared. Our results show that cathepsin D facilitates the TRAIL-induced apoptosis in enzymatic activity-dependent manner. We investigated the role of caspases and the Bcl-2 protein family in this process. The increased activation of the initiator caspase 8 mediating the direct activation of effector caspase 3 emerged as a molecular mechanism of the cathepsin D–mediated pro-apoptotic effect, whereas the activated Bid-mediated destabilisation of mitochondria acts as a latter amplification of the apoptotic process. Both TRAIL-induced apoptosis and cathepsin D–mediated pro-apoptotic effect are regulated by endolysosomal compartment acidification. We also addressed the question of potential application and effectivity of the specific cathepsin D inhibitors in cancer cells. The results of this study provided insight into the function of the lysosomal cathepsin D protease in the TRAIL-induced apoptosis and identified the cathepsin D enzymatic activity as a factor affecting sensitivity of breast cancer cells to TRAIL.
