Differentiation of human embryonic stem cells into odontoblasts

Abstract:

Během embryonálního vývoje podstupují buňky progresivní specializaci. Analýzy buněčných embryonálních linií ukázaly, že proces postupné diferenciace probíhá stupňovitým způsobem. Dentální mesenchymální buňky jsou příkladem buněčné linie, která během embryonálního vývoje podstupuje zmiňovanou progresivní determinaci. Tyto buňky formují komplexní morfologii zubu díky interakcím s epiteliální tkání (odvozena ze stomodeálního ektodermu) a na konci celého procesu se diferencují do několika specializovaných buněčných typů, z nichž každý buněčný typ dále produkuje tkáňově specifickou extracelulární matrix. Linie dentálních mesenchymálních buněk, kterou označujeme jako ektomesenchym, je odvozena z buněk kraniální neurální lišty a po determinaci a následné diferenciaci vytváří všechny struktury zubu a jejich podpůrné tkáně s výjimkou skloviny. Ektoderm a ektomesenchym jsou tak tkáněmi, které řídí tvorbu zubu od její iniciace, přes morfogenezi až k cytodiferenciaci. Mezi hlavní regulátory řídící tyto procesy patří fibroblastové růstové faktory, především však FGF8. Tento růstový faktor je začleněn mezi epiteliální signály, které reprimují expresi genu Hoxa2 během formování prvního žaberního oblouku. FGF8 hraje také klíčovou roli během procesu, kdy buňky kranio-neurální lišty získávají schopnost se diferencovat v odontoblasty a tvořit zub. Bohužel účinnost komerčně dostupných růstových faktorů pro laboratorní účely není v současné době stále stroprocentně garantovaná a může se také lišit v závislosti na bakteriálním kmeni, ve kterém byl protein produkován. Z tohoto důvodu jsem se ještě předtím, než jsem začala studovat diferenciaci lidských ES buněk a neurálních prekurzorů, pokusila o stabilní produkci zmiňovaného růstového faktoru. Tato práce přispívá ke studiu odontogeneze, které může napomoci porozumění řadě obecných aspektů embryogeneze a naopak znalosti o morfogenetických mechanismech vývoje jiných epitelio-mezenchymálních orgánů přispívají k objasňování odontogeneze. Práce se zaměřuje na vytvoření vhodných podmínek pro diferenciace buněk neurální lišty do odontogenní linie, výrazným způsobem přispěla k rozpracování technologie přípravy fibroblastového růstového faktoru 8 a jeho stabilní produkci v naší laboratoři. Poskytuje první reálný základ pro diferenciaci prekurzorů buněk neurální lišty do odontogenní linie.

Abstract:

During embryonic development cells undergo progressive specialization. Analyses of embryonic stem cell lines showed that the process of progressive differentiation is stepped manner. Dental mesenchymal cells are a cell line which undergo a progressive determination during embryonic development. These cells form morphology of the tooth due to interactions with epithelial tissue (derived from ectoderm) and they differentiate into several specialized cell types at the end of the process, each cell type also produces tissue-specific extracellular matrix. A line of dental mesenchymal cells, which are known as ektomesenchym is derived from cranial neural crest cells and the determination and subsequent differentiation creates all the structures of the tooth and its supporting tissues except the enamel. Ectoderm and ektomesenchym are tissues that control the developing of a tooth from its initiation through morphogenesis to cytodiferenciaci. The main regulators of these processes include group of fibroblast growth factors, primarily FGF8. This growth factor is incorporated within the epithelial signals and repress Hoxa2 gene expression during the formation of the first branchial arch. FGF8 plays a key role during the process where cells cranio-neural crest acquire the ability to differentiate into odontoblasty and tooth form. Unfortunately, the effectiveness of commercially available growth factors for laboratory purposes is not currently in a one-hundred-percent guaranteed, and may also vary depending on the bacterial strain in which the protein is produced. For this reason before I started studying the differentiation of human ES cells and neural precursors, I attemp to stable production of FGF8. This work contributes to the study of odontogenesis, which can help general understanding of many aspects of embryogenesis and contrast knowledge about mechanisms of morphogenetic development of mesenchymal-epithelial cells of other organs contribute to the clarification of odontogenesis. The work focuses on creating appropriate conditions for differentiation of neural crest cells in odontogenic lines contributed significantly to the development of technology of fibroblast growth factor 8 and stable production in our laboratory. Provides the first real basis for the differentiation of precursor cells in odontogenic neural crest lineage.
