Influence of pH on binding properties of lectins

Abstract:

Tato práce se zabývá vlivem pH na vazebné vlastnosti lektinů. Strukturní a funkční studie bakteriálních lektinů jsou v poslední době objektem zájmu, protože právě tyto proteiny se schopností vázat sacharidy umožňují patogenům připojit se na hostitelskou buňku a následně ji infikovat. Práce se věnuje lektinu RSL z bakterie Ralstonia solanacearum. Tato bakterie se vyskytuje po celém světě a způsobuje vadnutí zemědělských plodin, což má za následek ekonomické ztráty především v tropickém klimatu. Bakteriální lektiny se navíc v přírodě můžou vyskytovat při jiném pH, než se běžně používá pří práci s proteiny v laboratoři. Pomocí izotermální titrační kalorimetrie, nano diferenční skenovací fluorimetrie a rezonance povrchového plasmonu byl zkoumán vliv pH na stabilitu a vazebné vlastnosti tohoto lektinu. Bylo zjištěno, že RSL je velmi stabilní lektin, který preferuje spíše kyselé pH a jeho vazebné vlastnosti úzce souvisí se stabilitou. Během těchto experimentů byl otestován „buffer screen“, což je sada 48 roztoků o různém pH, iontové síle a přídavku aditiv. „Buffer screen“ se ukázal býti vhodným pro použití na různých „high-throughput“ biofyzikálních metodách při studiu lektinů.

Abstract:

This thesis deals with the influence of pH on binding properties of lectin. Structural and functional studies of bacterial lectins have recently been of interest because these carbohydrate-binding proteins allow pathogen to attach to the host cell and subsequently infect it. The thesis is devoted to lectin RSL from bacterium Ralstonia solanacearum. This bacterium is distributed worldwide and causes lethal wilt in agricultural crops, resulting in economic losses mainly in tropical climates. In addition, bacterial lectins can occur in nature at pH which is different from that usually used when working with proteins in the lab. The influence of pH on stability and binding properties of this lectin was investigated using isothermal titration calorimetry, nano differential scanning fluorimetry and surface plasmon resonance. It has been found that RSL is very stable lectin that prefers acidic pH and its binding properties are closely related to stability. During these experiments, a set of 48 conditions of various pH, ionic strength and additives called “buffer screen” was tested and proved to be suitable for use in various high-throughput biophysical methods in the studies of lectins.
