Staphylococcal enterotoxin determination by means of mass spectrometry

Abstract:

Úvod: Otrava vyvolaná požitím potravy kontaminované stafylokoky je jedna z nejčastějších typů onemocnění souvisejících s příjmem potravy. Onemocnění je způsobeno stafylokokovými enterotoxiny (SEs) přítomnými v potravinách jako např. v mléčných produktech, mase či vejcích. SEs jsou proteiny rozpustné ve vodě o nízké molekulové hmotnosti uvolňované do média během pěstování určitých druhů gram-pozitivních bakterií Staphylococcus aureus. Tento patogen je příčinou mnoha infekcí v lidském organismu. Je vyvíjena snaha o nalezení vhodné a citlivé metody detekce stafylokokových enterotoxinů na úrovni nižší, než je toxicita těchto proteinů (přibližně 100 ng). Metody: V této práci jsme zjišťovali přítomnost enterotoxinu H a B pomocí kombinace separační metody (OFFGEL frakcionace, 1D SDS-PAGE, GPC, RP-HPLC) a následné detekce a stanovení toxinu hmotnostní spektrometrií. Výsledky a závěr: Enterotoxiny byly identifikovány pouze po separaci technikou jednorozměrné elektroforézy. Poněvadž dosavadní výsledky jsou neuspokojivé, budeme testovat další separační metody pro následnou analýzu komplexní proteinové směsi obsahující enterotoxiny hmotnostní spektrometrií až do dosažení vhodných metod pro jejich kvantifikaci.

Abstract:

Background: Staphylococcus food poisoning is one of the most common food-borne diseases resulting from the ingestion of staphylococcal enterotoxins (SEs) in food matrices, like diary product, meat products and egg mixtures. SEs are small molecular weight water soluble, simple proteins released into the medium during growth by certain strains of the gram-positive bacterium, Staphylococcus aureus. This pathogen causes a variety of infections in humans. The significant effort is focused on finding reliable and sensitive method for the detection of staphylococcal enterotoxins at a lower threshold than the toxicity level (about 100 ng). Methods: We investigated the presence of enterotoxin H and B by using a combination of separation methods (OFFGEL fractionation, 1D SDS-PAGE, GPC, RP-HPLC) and subsequent detection and toxin determination by mass spectrometry. Results and conclusion: Enterotoxins were identified only by one-dimensional electrophoresis separation technique. Since the results are unsatisfactory, we will examine other separation methods for next analysis of the complex protein mixture containing enterotoxins by techniques of mass spectrometry to reveal the appropriate methods for their quantification.
