Anotace: HMGB1 a HMGB2 jsou evolučně vysoce konzervované proteiny, které se vážou s DNA a zvyšují přístup transkripčním proteinům ke specifickým oblastem na DNA. HMGB1/2 se účastní celé řady buněčných procesů včetně replikace, transkripce, opravy DNA, proliferace, diferenciace, apoptózy a růstu nádorů. Doposud nebylo známo, zda se tyto proteiny účastní regulace typických funkcí lidských embryonálních kmenových buněk (hESCs) nebo jejich diferencovaných derivátů. Inhibice exprese proteinů HMGB1/2 vede u eukaryotních buněk ke zvýšení citlivosti buněk k protinádorovým léčivům. Není však známo jaký je přesně mechanismus účinku těchto léčiv u hESCs a jejich diferencovaných derivátů jako jsou například neuroektodermální kmenové buňky (hNECs). V předkládané dizertaci jsme použili indukovatelné a stabilně transfekované hESCs, které jsou schopné inhibovat expresi proteinů HMGB1 a/nebo HMGB2 („knock-down“ vlivem shRNA). Prokázali jsme, že inhibice exprese HMGB1 a/nebo HMGB2 neovlivňuje klíčovou vlastnost lidských embryonálních kmenových buněk, pluripotenci, a má pouze minimální vliv na rychlost proliferace, buněčný cyklus a apoptózu. Diferenciace těchto buněk na neuroektodermální buňky naopak vede k dramatickým změnám v rychlosti proliferace, k indukci apoptózy, změnám v telomerázové aktivitě a účinnosti diferenciace. Je velmi zajímavé, že tyto biologické procesy jsou ovlivnitelné pouze tehdy, když dochází k inhibici pouze jednoho ze dvou proteinů HMGB. Simultánní inhibice exprese HMGB1 a HMGB2 naopak vykazuje znaky normálního fenotypu. Dále jsme prokázali, že protinádorové léčivo etoposid sice indukuje rychlou a rozsáhlou apoptózu v jak nediferencovaných hESCs, tak v diferencovaných buňkách (hNECs), diferencované buňky jsou ale výrazně citlivější k tomuto léčivu. Předkládaná disertace rozšiřuje naše poznatky o regulaci diferenciace hESCs a lidských neuroektodermálních buněk a podává zcela nové informace o odlišné funkci proteinů HMGB1 a HMGB2 v počátečních stádiích lidského vývoje.

Abstract: The HMGB1 and HMGB2 are evolutionarily highly conserved non-histone nuclear proteins that bind to DNA and elevate access to transcriptional proteins on specific targets. In addition, these chromatin binding proteins have been involved in numerous cellular processes, including replication, regulation of gene transcription, DNA repair, proliferation, differentiation, apoptosis, and tumor growth. It is unclear whether they are implicated in regulating the typical functions of pluripotent human embryonic stem cells (hESCs) and/or those of the differentiated derivatives of hESCs. In eukaryotic cells, lacking of HMGBs in many malignancies considerably enhances drug sensitivity to anticancer agents for instance etoposide, suggesting their wide role in improvement of current chemotherapeutic treatments. However, the specific mechanisms governing the biological activities of HMGB proteins in etoposide-treated hESCs and differentiated human neuroectodermal cells (hNECs) remain unknown. In this study we have used inducible stably transfected hESCs capable of shRNA-mediated knock-down (KD) of HMGB1 and HMGB2. We demonstrated that down-regulation of HMGB1 and/or HMGB2 in pluripotent hESCs has not impact on stemness and induces only negligible changes to the proliferation rate, cell cycle profile, and apoptosis. After induction of differentiation, the effect of down-regulating HMGB proteins on cell proliferation, apoptosis, telomerase activity, and the efficiency of differentiation towards neuroectodermal lineage becomes manifested. Importantly, all of those processes are affected only when one, but not both, of the two proteins is downregulated; as simultaneously HMGB1 and HMGB2 KD led to normal phenotype. Treatment of hESCs and hNECs with etoposide could induce a rapid and extensive induction of apoptosis in both undifferentiated hESCs and differentiated hNECs upon silencing of HMGB1 and /or HMGB2,with the sensitivity being more profound in differentiated hNECs. Thus, the thesis not only advances the knowledge about the regulation of differentiation of hESCs and human neuroectodermal cells but also brings a novel finding of the distinct roles of HMGB1 and HMGB2 proteins during early human development.
