Molecular phylogeny of the infraorder Bibionomorpha (Diptera), with a main focus on the family Mycetophilidae

Abstract:

The species-rich family Mycetophilidae (fungus gnats) belongs to the megadiverse insect infraorder Bibionomorpha (Diptera). The diversity of these groups provides an extensive platform for the study of their taxonomy and phylogeny. There have been many studies published on this issue, but there still remain a lot of controversial topics to be discussed. This is often due to insufficient taxon sampling used for the reconstructions of their phylogeny or other missing data, either morphological or molecular. The aim of this work is to summarize the current knowledge about the phylogeny of Bibionomorpha, with focus on the family Mycetophilidae, and to carry out a new phylogenetic analysis based on a wide range of species and molecular data that should solve or help to solve problematic issues related to the phylogenetic relationships of these diverse groups at various levels. Another aim is to describe some of the unknown species from the family Mycetophilidae. Specimens were collected in various zoogeographical regions using Malaise traps or sweep netting, or some of them were obtained from museum collections. The molecular phylogenies were reconstructed based on mitochondrial (12S, 16S, COI, COII, cytB) and nuclear (18S, 28S, ITS2, CAD, MCS) gene markers. Within the infraorder Bibionomorpha, the monophyly of Bibinomorpha sensu lato and sensu stricto was established with high support. The monophyly of the superfamily Sciaroidea and interrelationships within this superfamily were also demonstrated, with mostly high support. The position of some Sciaroidea incertae sedis genera was also clarified. The relationships among all subfamilies of Mycetophilidae were interpreted and attention was paid mainly to the subfamilies Manotinae, Leiinae, Mycomyinae and Gnoristinae (including Metanepsiini). In this family, the position of several enigmatic genera has also been elucidated and four new species were described.

Abstract:

Čeleď Mycetophilidae (bedlobytkovití) patří do druhově bohatého infrařádu Bibionomorpha (Diptera). Druhová rozmanitost této skupiny poskytuje rozsáhlou platformu pro studium taxonomie a fylogeneze těchto skupin. Problematice taxonomie a fylogeneze se věnovalo mnoho studií, navzdory tomu nám stále zůstává mnoho nezodpovězených otázek a sporných výsledků. To je často způsobeno nedostatečným počtem taxonů použitých pro rekonstrukci fylogeneze nebo chybějícími morfologickými či molekulárními daty. Cílem dizertační práce je shrnout dosavadní poznatky o fylogenezi infrařádu Bibionomorpha se zaměřením na čeleď Mycetophilidae a provést nové fylogenetické analýzy na základě širokého spektra druhů a jejich molekulárních dat, které pomohou vyřešit problematické fylogenetické vztahy v rámci těchto diverzních skupin. Dalším cílem je popsat vybrané nové druhy z čeledi Mycetophilidae. Sběr jedinců probíhal v různých zoogeografických oblastech pomocí Malaiseho pastí a smýkání, popřípadě bylo využito muzejních exemplářů. Rekonstrukce molekulární fylogeneze byly provedeny na základě mitochondriálních (12S, 16S, COI, COII, cytB) a jaderných (18S, 28S, ITS2, CAD, MCS) genetických markerů. S vysokou statistickou podporou byla prokázána monofyletičnost Bibionomorpha sensu lato, sensu stricto a nadčeledi Sciaroidea. Rovněž byly demonstrovány vztahy uvnitř nadčeledi Sciaroidea. Byla vyřešena pozice některých rodů doposud zařazených mezi Sciaroidea incertae sedis. Byly předloženy výsledky analýz vztahů mezi podčeleděmi uvnitř čeledi Mycetophilidae a pozornost byla zaměřena především na podčeledi Manotinae, Leiinae, Mycomyinae a Gnoristinae (včetně tribu Metanepsiini). V těchto podčeledích byly vyřešeny sporné pozice některých taxonů a také byly popsány 4 nové druhy.
