Immunogenomic dissection of post-vaccination antibody immune response in horses

Abstract:

The aim of the diploma thesis was to identify the the genes and their polymorphisms associated with high and low response against horse herpesvirus 1 vaccination within old kladruber horse. With this goal the methodologies of their genotypization were worked out, Obtained markers can be used in other association studies. The implemented markers considered to be the candidate genes were used as well within old Kladruby horses (some interleukin representatives and MXA). Specific primers were used to identify the SNP of the IRF3, IRF7 and MYD88 genes and the genotypization methods for IRF7 and MYD88 genes were worked out. SNP were foung for IRF3 gene in 3´UTR area, for IRF7 gene in intronic area and for MYD88 gene in exonic area. The amplification of TRIF gene was not successful. It was impossible to sequence the TLR9 gene and no polymorphism was discovered with TLR7 gene. Based on sequences that were found, according the PCR-RFLP method, the methodology was worked out and implemented. The associations were discovered between SNP in some candidate genes and individual variability in post-vaccination antibody imine response against EHV1 (mainly IL-4R gene). Based on these results, the research on the role of some of these in genetic determination of immune response against EHV1 will be carried on.

Abstract:

Cieľom diplomovej práce bola identifikácia génov a ich polymorfizmov, ktoré sú asociované s vysokou a nízkou odpovedavosťou na vakcináciu proti konskému herpesvírusu 1 u starokladrubského koňa. S týmto cieľom boli vypracované metódy ich genotypizácie. Získané markery sú využiteľné v ďalších asociačných štúdiách. Tu boli použité u starokladrubských koní s už zavedenými markermi ďalších kandidátnych génov (niektorých interleukínov a génu MXA). Za pomoci použitia špecifických primerov boli u génov IRF3, IRF7 a MYD88 nájdené SNP a pri IRF7 a MYD88 vypracované metodiky genotypizácie. U génu IRF3 v oblasti 3´UTR, u génu IRF7 v intrónovej a u MYD88 v exónovej oblasti. Gén TRIF sa nepodarilo amplifikovať, gén TLR9 nebolo možné nasekvenovať a u génu TLR7 nebol nájdený polymorfizmus. Na základe zistených sekvencií bola vypracovaná metodika pomocou metódy PCR-RFLP a následne zavedená do praxe. Boli zistené asociácie medzi SNP v niektorých kandidátnych génoch a individuálnou variabilitou v postvakcinačnej protilátkovej odpovede proti vírusu EHV1 (predovšetkým gén IL-4R). Úloha niektorých z nich v genetickej determinácii imunitnej odpovedavosti proti vírusu EHV1 bude na základe týchto výsledkov naďalej skúmaná.
