An amperometric biosensor for the determination of ethanol in selected samples of alcoholic drinks

Abstract:

Cílem této diplomové práce bylo navrhnout a zkonstruovat amperometrický biosenzor určený ke stanovení ethanolu ve vybraných vzorcích nápojů s obsahem alkoholu. V teoretické části jsou detailně popsány jednotlivé komponenty utvářející výsledný biosenzor a možné postupy určené k jeho zhotovení. Následuje samostatná část věnovaná vybraným postupům ke stanovení ethanolu, ať už jde o tradiční alkoholové testy pro měření v terénu či o moderní laboratorní metody využívající sofistikovanou instrumentální techniku. Experimentální část se zabývá výběrem vhodného amperometrického převodníku a způsoby imobilizace jednotlivých součástí konfigurace testovaného biosenzoru, tj. enzymu, kofaktoru a mediátoru; to vše s následnou optimalizací klíčových pracovních podmínek a přístrojových parametrů. Konečná konfigurace pak byla ověřována na několika typech alkoholických nápojů, kdy získané údaje posloužily k definování možností biosenzoru a jeho uplatnění v analýze reálných vzorků.

Abstract:

The aim of this diploma thesis was to design and construct an amperometric biosensor intended for the determination of ethanol in selected samples containing alcohol. Theoretical part deals in detail with the main components of amperometric biosensors and especially those applicable to the ethanol sensing. Then, a special section is devoted to the manufacturing procedures of such biosensors, followed by a survey of traditional alcohol testers and modern instrumental methods for the determination of ethanol. In the experimental part, of main interest were amperometric transducers and the corresponding procedures for immobilization of enzyme, cofactor, and mediator, together with optimisation of the key experimental and instrumental parameters. The final configuration has been examined on various alcoholic beverages, allowing us to obtain a number of useful data for defining the overall performance of the biosensor with respect to its use in analysis of real samples.
