Preparation and chemical modification of silver particle layers on glass substrates for applications in surface enhanced Raman spectroscopy

Abstract:

Cílem diplomové práce byla příprava a chemická modifikace vrstev koloidních částic stříbra na skleněném podkladu za účelem jejich aplikace v oblasti povrchem zesílené Ramanovy spektroskopie. Nanočástice stříbra o průměrné velikosti 28 nm byly připraveny redukcí amoniakálního komplexu stříbrných iontů D-maltosou. Připravená disperze nanočástic stříbra byla následně využita na přípravu vrstev nanočástic stříbra za použití poly(diallyldimethylamonium chloridu). Rovněž byly stříbrné vrstvy připravovány pomocí sonochemické metody. Efektivita připravených vrstev v rámci jejich aplikovatelnosti pro účely povrchem zesílené Ramanovy spektroskopie byla testována za využití roztoku adeninu s finální koncentrací roztoku 10-5 mol?dm-3. Připravená vrstva nanočástic stříbra za použití PDDA nevykazovala dostatečnou homogenitu, zatímco stříbrné vrstvy připravené pomocí sonochemické metody byly schopny efektivně zesilovat Ramanův signál adeninu při použití laseru s excitační vlnovou délkou 785 nm a zároveň poskytovaly potřebnou homogenitu. Tento fakt byl velice žádoucí pro následnou chemickou modifikaci stříbrné vrstvy. Chemická modifikace vrstev koloidních částic stříbra byla prováděna pomocí linkerů o složení aminokyselina-amin. K aktivaci karboxylových skupin pro následné navázání, co největšího množství Streptavidinu byla využita aktivační činidla (1-[3-(dimethylamino)propyl]-3-ethylcarbodiimide methiodide (EDC), N-hydroxysuccinimide sodium (NHS)).Na stříbrnou vrstvu s navázaným Streptavidinem byl přes biotinylovanou protilátku vhodnou pro daný protein imobilizován Protein A, který je typický pro bakterie Staphylococcus Aureus. Imobilizace TAU proteinu na stříbrnou vrstvu s navázaným Streptavidinem byla provedena přes biotinylovanou protilátku pro TAU protein, který je typickým markerem Alzheimerovy choroby.

Abstract:

The aim of this diploma thesis was the preparation and chemical modification of layers of colloidal silver particals on the glass substrate for the purpose of the application in the surface-enhanced Raman spectroscopy. The silver nanoparticles with an average size 28 nm were prepared via the reduction of ammonia complex of silver ions by D-maltose. The concentration of thus prepared silver nanoparticals was 108 mg?dm-3. Consequently the prepared dispersion of silver nanoparticles was used to prepare layers of silver nanoparticles while using the poly(diallyldimethylamonium chloride). There were also some silver layers, which were prepared via the sonochemical method. The effectiveness of the prepared layers was tested in the area of the surface-enhanced Raman spectroscopy. The solution of the adenine with the final concentration 10-5 mol?dm-3 was used for these tests. The layer of silver nanoparticles, which was prepared while using the PDDA, did not provide the required homogeneity. However, the silver layers (prepared via the sonochemical method) were able to amplify the Raman signal of the adenine effectively while the laser with the excitation wavelength 785 nm was used. These layers also provided the required homogeneity. This fact was very desirable for the further chemical modification of the silver layer. The chemical modification of the layers of colloidal silver particles was done while using linkers. The composition of these linkers was amino acidamine. The activating reagents (EDC, NHS) were used to activate the carboxyl groups to establish as much streptavidine as possible. The protein A was immobilized on the silver layer with established streptavidine via biotynaled antibody. This antibody was suitable for the given protein. Protein A could be further used to detect the bacteria Staphylococcus Aureus. The immobilization of the TAU protein on the silver layer with established streptavidine was done via the biotynaled antibody for the TAU protein. TAU protein is possible to use as a marker for the Alzheimer's disease.
