Response and adaptation of Samonella to stress caused by interaction with the external environment

Abstract:

Bacteria of the genus Salmonella are the major enteric pathogens causing disease in humans and animals. Bacteria proliferate in the gastrointestinal tract of warm-blooded and cold-blooded vertebrates and successfully survive in the external environment (wastewater, river, sea water or foodstuff). Therefore, during the evolution Salmonella had to develop defense mechanisms that enable quick adaptation to changing conditions. Information on these mechanisms can be used to propose proceedings to prevent the spread of bacteria during food processing or for the preparation of vaccine strains. RpoS protein is the master regulator of the general stress response. RpoS is the sigma subunit of RNA polymerase, which regulates the transcription of more than 500 genes. These genes are encoding proteins that are involved in protection of the bacteria against many stress factors and arrange the entry of bacterial culture in the stationary phase of growth. In the laboratory where I process my work, eight genes of unknown function were identified using the microarray analysis (ymdF, STM1513, yciG, yciE, yciF, yliH, ybgS and yeaG) that are over expressed during the late stationary phase of growth. All these genes are transcribed under the controle of RpoS. SE147 Δ8G strain lacking all these 8 genes was prepared to study functions of the genes. SE147 Δ8G was exposed to a great variety of stress factors however any difference when compared with wild type strain was observed. As a response to the deteriorating conditions when bacterial culture entry in the stationary phase the permeability of the cell membrane is reduced. The rate of uptake of gentian violet into the bacterial cytoplasm corresponds with the rate of bacterial membrane permeability for small organic molecules. Therefore, I compared strain SE147 Δ8G with wild-type strain SE147 for the ability of the gentian violet uptake. I proved, that the rate of uptake of gentiane violet is higher in the strain SE147 Δ8G cultured in M9 minimal medium. This is the first difference in phenotype between SE147 Δ8G and wild type strain so far identified. Based on this result I can predict that one or more of the studied genes is involved in the reduction of membrane permeability in the late stationary phase of growth accompanied by starvation.

Baktérie rodu Salmonella patria k významným črevným patogénom spôsobujúcim ochorenia u ľudí a zvierat. Pomnožujú sa v gastrointestinálnom trakte teplokrvných a studenokrvných stavovcov a úspešne prežívajú v vonkajšom prostredí (odpadnej, riečnej, morskej vode i v potravinách). Aby salmonely dokázali prežívať v takto rozdielnych prostrediach, museli sa u nich behom evolúcie vyvinúť obranné mechanizmy, ktoré im umožňujú rýchlo reagovať na zmeny podmienok. Štúdium týchto obranných mechanizmov môže byť využité pri tvorbe opatrení brániacich šíreniu baktérií behom zpracovávania potravín a taktiež pri príprave vakcinačných kmeňov. Hlavným regulátorom obecnej odpovedi na stres je proteín RpoS. Je to sigma podjednotka RNA polymerázy, ktorá reguluje transkripciu viac než 500 génov. Jedná sa o gény kodujúce proteíny, ktoré slúžia k ochrane baktérií pred mnohými stresovými faktormi a ďalej sprostredkovávajú vstup bakteriálnej kultúry do stacionárnej fázy rastu. Pomocou microarray analýzy bolo na pracovisku, na ktorom zpracovávam svoju bakalársku prácu identifikovaných 8 génov o neznámej funkcii (ymdF, STM1513, yciG, yciE, yciF, yliH, ybgS a yeaG), ktorých transkripcia prebieha pod kotrolou RpoS a niekoľkonásobne sa zvyšuje po vstupe bakteriálnej kultúry do neskorej stacionárnej fázy. Za účelom štúdia funkcie týchto génov bol skonštruovaný kmeň SE147 Δ8G s deleciou všetkých 8 génov zároveň. Tento kmeň bol otestovaný na zmenu odolnosti k mnohým stresovým faktorom, ale žiadny rozdiel pri porovnávaní s kmeňom divokého typu nebol pozorovaný. Na zhoršujúce sa podmienky pri vstupe bakteriálnej kultúry do stacionárnej fázy rastu baktéria reaguje znížením permeability bunkovej membrány. Schopnosť baktérií pohlcovať genciánovú violeť vypovedá o priepustnosti bakteriálnej membrány pre malé organické molekuly, preto som túto vlastnosť porovnala u SE147 Δ8G v porovnaní s kmeňom SE147 divokého typu. Podarilo sa mi prekázať, že kmeň SE147 Δ8G vykazuje vyššiu priepustnosť membrány pre genciánovú violeť po kultivácii v minimálnom médiu M9, čo je prvý pozorovaný fenotypový prejav tohto kmeňu pri porovnaní s kmeňom divokého typu. Na základe tohoto výsledku môžem predpovedať, že jeden alebo viacej zo študovaných génov sa podiela na znížení permeability membrány v neskorej stacionárnej fázy rastu doprevádzanej hladovaním.
