Stress priming and memory in plants

Abstract:

Aby mohly rostliny odolávat opakujícím se vnějším stresovým vlivům, vyvinula se u nich schopnost aklimatizace. Pokud je rostlina ještě před samotným stresem, který by mohl být letální či rostlinu vážně poškodit, vystavena stresu mírnějšímu, bude připravena reagovat efektivněji a dokáže se lépe se stresovým vlivem vyrovnat. Tento proces přípravy na následující stres je označován pojmem priming. Tato práce stručně popisuje mechanismy tohoto fenoménu a různé možnosti navození adaptace. V praktické části byl vytvořen modelový systém takto adaptovaných rostlin Arabidopsis thaliana vůči tepelnému stresu. Rostliny, které prošly procesem primingu vykazovaly po následném stresovém tepelném ošetření větší vzrůst, vyšší hmotnost, vyšší obsah chlorofylu a celkově lepší vitalitu než rostliny, které byly stresovému ošetření vystaveny bez předchozího procesu primingu. Dále byla provedena proteomická analýza, která se zaměřila na proteiny, jejichž množství bylo během primingu signifikantně změněno. Tato analýza ukazuje mimo jiné výrazné interakce sledovaných proteinů, což nasvědčuje, že zodpovídají za společnou reakci na proteinové úrovni.

Abstract:

To deal with recurring environmental stress factors, plants have developed the ability to acclimate. If the plant is exposed to milder stress before the lethal or damaging stress occurs, it will be prepared to react more effectively and cope better with the stress factor. This process of preparing for the subsequent stress is referred to as priming. This bachelor thesis briefly describes the mechanisms of this phenomenon and various possibilities of inducing adaptation. In the experimental part model system of heat stress adapted plants Arabidopsis thaliana was created. Plants that were primed showed greater growth, higher weight, higher chlorophyll content and overall better vitality compared to plants that were exposed to the heat stress without previous priming. Furthermore, proteomics analysis was performed, which was focused on proteins, whose amount was significantly changed during priming. This analysis shows significant interactions of the studied proteins, which suggests that they are responsible for the concerted response at the protein level.
