The role of plant hormones for efficient fertilization processes at high temperatures in Arabidopsis and oilseed rape

Abstract:

Práce se zabývá vlivem vysokých teplot na fertilizační procesy rostlin. V teoretické části jsou uvedeny základní fytohormony, zvýšená pozornost se věnuje auxinu, jeho syntéze a genům, které ho kóduji. Dále je popsána morfologie reprodukčních orgánů, vliv vysokých teplot na rostliny a jsou zde popsány klíčové principy molekulárně biologických metod, které byly v této práci použity. V praktické části byl vytvořen vektorový plazmid a transformován do rostlin. Detailně prostudovány různé části reprodukčního procesu, jakož je vývoj pylu, růst pylové láčky, vývoj pestíků a semen v různých teplotních podmínkách. Klíčovými rostlinami pro experimenty byla Arabidopsis thaliana (wildtypy a mutantní linie) a kultivary řepky. V závěru práce je vyhodnocení získaných dat a srovnání s dostupnou literaturou.

Abstract:

The thesis deals with the influence of high temperatures on fertilization processes of plants. In the theoretical part, the basic phytohormones are mentioned, attention is paid to auxin, its synthesis and genes that encode the enzymes involved in it. Furthermore, the morphology of reproductive organs, the effect of high temperatures on plants and the key principles of molecular biological methods used in this work are described. In the practical part, a vector plasmid was created and transformed into plants. The various parts of the reproductive process, such as the development of pollen, the growth of pollen, the development of pistils and seeds in various temperature conditions, have been studied in details. Key plants for the experiments were Arabidopsis thaliana (wild types and mutant lines) and rape cultivars. The work is based on the evaluation of acquired data and comparison with available literature.
