Characteristics of morphological and functional parameters of bovine oocytes

Abstract:

Meiotická a vývojová kompetence patří mezi základní charakteristiky určující kvalitu bovinních oocytů. Cílem disertační práce bylo charakterizovat bovinní oocyty s odlišnou meiotickou kompetencí a stupněm zdraví z hlediska morfologických a funkčních vlastností jejich mitochondrií a definovat mitochondriální změny, ke kterým dochází během meiotického zrání a fertilizace v podmínkách in vitro. Oocyty s vyšší a nižší meiotickou kompetencí byly získány separovaně buď ze středních nebo malých antrálních folikulů z ovarií jatečných krav v definovaném stádiu folikulogeneze. Na základě morfologického hodnocení kumulárních buněk a cytoplasmy byly oocyty selektovány na zdravé, lehce a středně atretické. Kategorie oocytů zrály a byly oplozeny podle standardních protokolů. V intervalech 0, 3, 7, 16 a 24 h po zrání nebo 6, 12 a 18 h po fertilizaci a vizualizaci mitochondrií a endoplazmatického retikula (ER), byly oocyty vyšetřeny konfokální mikroskopií. Obsah ATP v cytoplazmě oocytů byl stanoven metodou chemiluminometrie. Byly nalezeny rozdíly v morfologických a funkčních vlastnostech mitochondrií u oocytů s vyšší a nižší meiotickou kompetencí před a po zrání. Zdravé více kompetentní oocyty aktivovaly mitochondrie během zrání dvakrát, ve stádiu GVBD a dosažení MII, zatímco méně kompetentní oocyty aktivovaly mitochondrie až ve stádiu kompletace meiózy. Zdravé více kompetentní oocyty formovaly mitochondriální klastry a produkovaly ATP po celou dobu zrání, zatímco lehce atretické více kompetentní oocyty a zdravé méně kompetentní oocyty ukončily tyto aktivity již v době nástupu MII. Ve srovnání se zdravými oocyty byly lehce atretické oocyty charakteristické akcelerací znovuzahájení meiózy, které korelovalo s nárůstem podílu oocytů s mitochondriálními klastry. Bylo prokázáno, že fertilizace zralých oocytů vyvolala rychlý rozpad komplexů mitochondriálních klastrů s ER, přesun mitochondrií z periferie oocytu do centrálních oblastí ooplasmy a relokalizaci sítě endoplazmatického retikula pod oolemou. Pokud oocyty nebyly oplozeny, v přítomnosti spermií si uchovaly vysoké energetické rezervy až po dobu 12 hodin, zatímco při kultivaci oocytů pouze ve fertilizačním médiu došlo k rozpadu ER-mitochondriálních komplexů již za 6 hodin. Závěrem lze shrnout, že byly charakterizovány mitochondriální změny a nalezeny specifické rozdíly v morfologických a funkčních vlastnostech mitochondrií u bovinních oocytů s odlišnou meiotickou kompetencí a úrovní zdraví, které zrály a byly oplozeny v podmínkách in vitro.

Abstract:

Meiotic and developmental competence are basic determinants of bovine oocyte quality. The aim of this dissertation was to characterize bovine oocytes with different meiotic competence and health state in terms of morphological and functional parameters of their mitochondria and to define mitochondrial changes that occur during meiotic maturation and fertilization in vitro. Oocytes with greater and lesser meiotic competence were collected separately either from medium or small antral follicles present on ovaries of slaughtered cows in defined stage of folliculogenesis. On the basis of morphological evaluation of cumulus cells and oocyte cytoplasm they were classified as healthy, light atretic and mid-atretic. The categories of oocytes were matured and fertilized according to standard protocols. In the time intervals of 0, 3, 7, 16 and 24 hours after maturation or 6, 12 and 18 hours after fertilization and staining of vital mitochondria and immunofluorescent visualization of endoplasmic reticulum (ER), the oocytes were examined with confocal microscopy. Content of ATP in oocyte cytoplasm was determined by chemiluminometry. Differences in morphological and functional parameters of mitochondria were found between oocytes with greater and lesser meiotic competence before and after maturation. Activation of mitochondria occurred twice during maturation of oocytes with greater meiotic competence, at the GVBD stage and at MII completion, while in oocytes with lesser meiotic competence, mitochondria was activated only at the time of meiosis completion. Healthy oocytes with greater meiotic competence were characterized by formation of mitochondrial clusters and production of ATP throughout the maturation interval, while light atretic oocytes with greater meiotic competence and healthy oocytes with lesser meiotic competence terminated these activities at the time of MII onset. Compared to healthy oocytes, light atretic oocytes were characterized by accelerated resumption of meiosis, which correlated with increased proportion of oocytes with mitochondrial clusters. Results of the experiments also proved that fertilization of matured oocytes caused fast disintegration of mitochondria-ER complexes, migration of mitochondria from the oocyte periphery to central areas of ooplasm and relocalization of the ER network under oolema. If the oocytes were not fertilized, they maintained high energy reserves for up to 12 hours in presence of spermatozoa, while cultivation of oocytes in fertilization medium without spermatozoa lead to disintegration of mitochondria-ER complexes within 6 hours after insemination. It can be concluded that there were changes characterized and specific differences found in mitochondrial parameters in bovine oocytes with different meiotic competence and health state, which were matured and fertilized in in vitro conditions.
