Změny v časné produkci cytokinů u Schwannových buněk po indukci zánětu v podmínkách in vitro

Early cytokine production changes in Schwann cells after inflammatory stimuli in vitro

Abstract:

Úvod. Zánětlivá odpověď Schwannových buněk, která se vyvíjí po traumatickém poškození nebo v průběhu různých onemocnění periferních nervů, je doprovázena aktivací Schwannových buněk. Zánětlivá aktivace Schwannových buněk je indukována prostřednictvím Toll-like receptorů, může být spuštěna exogenními patologickými molekulami nebo endogenními ligandy produkovanými během Wallerovy degenerace. Mediátory, které vznikají během této aktivace, se účastní koordinace buněčných reakcí, urychlují komunikaci mezi buňkami a podílejí se na degeneraci a následné regeneraci poškozeného nervu. Metody. Změny v produkci sledovaných proteinů - NFκB, STAT3, IL-lβ a IL-6 byly měřeny pomocí imunocytochemie, western blotting, ELISA. Charakterizace buněk RT4 byla provedena průtokovou cytometrií. Výsledky. Disertační práce se zaměřila na časné zánětlivé reakce RT4 buněk, konkrétně na změny produkce proteinů TLR4, NFκB, IL-lβ, pSTAT3 a IL-6 po stimulaci LPS v podmínkách in vitro. Významně zvýšené hladiny proteinů NFκB, IL-lβ, pSTAT3 a IL-6 byly nalezeny 1 hodinu po působení LPS, což ukazuje na jejich zapojení do iniciace zánětlivé reakce Schwannových buněk. Tato iniciace byla indukována bez zvýšené exprese proteinu TLR4, ale byla doprovázena výskytem TLR4 v časných endozomech. Hladiny proteinu se během následujících 6 hodin stimulace pomocí LPS snížily. Produkce NFκB, IL-lβ a pSTAT3 po 24 hodinách stimulace LPS opětovně zvýšena. Naopak kontinuelní snižování IL-6 v průběhu času po ovlivnění LPS nebylo očekávané. Hladiny solubilního proteinu IL-6 v kultivačním médiu také klesaly, což korespondovalo se snižujícími se hladinami LPS během 24 hodinové stimulace buněk. Závěr. Výsledky našich in vitro experimentů prokázaly dynamiku časného zánětlivého profilování Schwannových buněk, ke kterému může také dojít in vivo aktivací TLR4 endogenními ligandy během WD nebo exogenními ligandy po poškození nervů.

Abstract:

Introduction. The inflammatory response of Schwann cells, which develops after the traumatic damage or within the course of various peripheral nerve diseases, is accompanied by activation of Schwann cells. The inflammatory activation of Schwann cells is induced by initiation via Toll-like receptors can be triggered by exogenous pathological molecules or endogenous ligands produced during Wallerian degeneration. The mediators arising during this activation are participating in coordination of the cells reactions, accelerate the cell-cell communication and participate in the degeneration and a subsequent regeneration of the damaged nerve. Methods. The changes in the production of monitored proteins – NFκB, STAT3, IL-1β and IL-6 were measured by Immunocytochemistry, Western blotting, ELISA. The characterization of RT4 cells was done with flow cytometry. Results. Dissertation thesis focused on the early period of inflammatory reactions by following the levels of TLR4, NFκB, IL-1β, pSTAT3, and IL-6 proteins after LPS treatment of RT4 cells under in vitro conditions. Significantly increased levels of NFκB, IL-1β, pSTAT3, and IL-6 proteins were found 1 h after LPS action indicating their involvement in the initiation of the inflammatory reaction of schwannoma cells. This initiation was induced without increased TLR4 protein expression, but was accompanied by the appearance of TLR4 in early endosomes. The protein levels decreased within the next 6 h of treatment with a subsequent increase of NFκB, IL-1β, and pSTAT3 after 24 h of LPS treatment. In contrast, continuous decrease of IL-6 over time following LPS treatment was unexpected. Levels of soluble IL-6 protein in the culture medium also decreased with decreasing levels of LPS over 24 h. Conclusions. The results of our in vitro experiments demonstrated a dynamic early inflammatory profiling of Schwann cells that can also occur in vivo via activation of TLR4 by endogenous ligands during WD or exogenous ligands after inflammatory nerve injury.
