Impact of physico-chemical properties of nanomaterials and exposure media on results of ecotoxicity tests

Abstract:

Prezentovaná disertační práce se zabývá problematikou testovaní ekotoxicity nanomaterialů a definovaním vyznamných fyzikalně-chemických parametrů nanočástic a expozičních medií, které tyto testy ovlivňují. Práce je rozdělena do tří svébytných celků, které vzájemně propojuje typické chování nanomaterialů, které je pro ně a jejich osud v životním prostředí charakteristické. Počáteční část práce je zaměřena na syntézu a charakterizaci základních fyzikalně-chemických parametrů stříbrných nanočástic (Ag NPs) a jejich osudu v kapalných expozičních médiích. K tomuto účelu byly využívány techniky dynamickeho rozptylu světla (DLS), spektroskopie ve viditelné a ultrafialové části spektra (UV-Vis), mikroskopie atomárních sil (AFM), elektronové mikroskopie (SEM), rentgenové difrakční analýzy (XRD), optické emisní spektrometrie s indukčně vazaným plazmatem (ICP - OES) a měření ?Ä-potenciálu koloidních systemů. Na charakterizační část navazují testy ekotoxicity Ag NPs ve vodných prostředích. K tomuto účelu byly použity vodní organismy hrotnatka velká (Daphnia magna), dánio pruhované (Danio rerio) a kapr obecný (Cyprinus carpio). Na nich bylo provedeno testování toxicity Ag NPs dle stávajících norem OECD, tak i za pomoci testů, které z norem OECD vycházejí, ale byly modifikovány s respektem ke zjištěným faktům z části charakterizační. Jelikož se aglomeračni chování jeví jako jeden z hlavnich problemů při testování ekotoxicity nanomateriálů ve vodných prostředích, zamýšlené modifikace měly za úkol snížit míru a rychlost aglomerace a získání monotónních závislostí dávka - účinek. Snaha potlačit aglomeraci nanočástic během terestrických testů vedla k využití agarového gelu jako kultivačního a expozičního média pro půdni organismy. Testování ekotoxicity bylo v tomto případě prováděno na roupici Enchytraeus crypticus a jako testovaný nanomateriál byly zvoleny nanočástice oxidu zinečnatého v práškové formě. Na nich byly ověřovány různé přístupy k provádění dispergace v expozičním médiu a na základě fyzikalně-chemické charakterizace vzorků bylo usuzováno na vztah mezi mírou aglomerace částic a jejich toxicitou pro pokusný organismus. Vyjma aglomerace jsou dalším procesem ovlivňujícím toxicitu nanočástic změny chemického složení jejich povrchu, které nastávají v důsledku interakce těchto částic se složkami životního prostředí. Další část práce je tedy věnována laboratorní modifikaci povrchu Ag NPs relevantní změně nanočástic v životním prostředí, jejich charakterizaci po přípravě i v expozičních médiích a buněčným testům. Ty byly provedeny na buňkách střevního epitelu pstruha duhového (Oncorhynchus mykiss) a byly koncipovány nejen jako porovnávací testy cytotoxicity, ale zároveň byly provedeny i experimenty schopností obou druhů těchto částic prostupovat skrz buněčné membrany do cytosolu a nasledně interagovat s důležitými buněčnými organelami.

Abstract:

The presented dissertation focuses on defining physico-chemical parameters of nanoparticles and exposure media that significantly affect the outcomes of ecotoxicological tests on nanomaterials. The dissertation is structured into three independent units that are all interconnected by the properties and behavior of nanomaterials that play a key role in their environmental fate. In the first unit, the synthesis and characterization of the basic physico-chemical parameters of silver nanoparticles (Ag NPs) and their behavior in liquid exposure media are discussed. The applied characterization techniques were dynamic light scattering (DLS), UltraViolet-Visible spectroscopy (UV-Vis), atomic force microscopy (AFM), electron microscopy (SEM), X-ray diffraction (XRD), optical emission spectrometry with inductively coupled plasma (ICP-OES) and ć-potential measurements of colloidal systems. The second unit then deals with the ecotoxicity tests of Ag NPs in aquatic environment. Ecotoxicity was tested on three aquatic organisms Daphnia magna, zebrafish (Danio rerio) and common carp (Cyprinus carpio) using OECD test methods as well as their modifications, which accounted for the observations from the characterization experiments. Since agglomeration appears to play a crucial role in nanomaterial ecotoxicity testing in aquatic environment. The applied modifications aimed to reduce the agglomeration rate and to obtain monotonic doseresponse curves. The effort to suppress the nanoparticle agglomeration during terrestrial ecotoxicity tests led to the application of agar gel as an exposure medium for terrestrial organisms. Toxicity of zinc oxide nanoparticles in powder form was tested on enchytraeids Enchytraeus crypticus and the influence of different dispersion techniques on the toxicity outcomes was examined. The evaluation of the relationship between the agglomeration rate and the toxicity for the target organism was bas based on the physico-chemical characterization of the samples of nanomaterials dispersed in the agar gel. Surface chemistry and the interactions with the components of the environment are another significant factor contributing to the complexity of nanomaterial ecotoxicity. Thus the third unit focuses on an environmentally relevant surface modification of Ag NPs and the characterization of the synthesized colloids as well as their dispersions in the exposure media for cytotoxicity tests. Comparative cytotoxicity experiments with native Ag NPs and their modified counterparts were conducted using intestinal epithelial cells of rainbow trout (Oncorhynchus mykiss). Penetration tests were performed to study the capability of the nanoparticles of both types to cross the cell membrane and thus interact with essential cellular organelles in the cytosol.
