Abstract:

Průkaz podmíněně patogenních mykobakterií ve vnějším prostředí a jejich zdravotní význam Mgr. Vladimír Beran Předložená disertační práce představuje souhrn publikací autora s komentáři k jednotlivým publikacím. Podmíněně patogenní druhy mykobakterií byly detekovány v mízních uzlinách prasat přikrmovaných neošetřenou i ošetřenou rašelinou. Použití rašeliny jako krmivového doplňku pro prasata může být z pohledu výskytu mykobakterií zdravotně rizikové. Prostředí akvárií stejně jako chované klinicky zdravé ryby obsahovaly velké množství mykobakterií, které tak mohou představovat zdravotní riziko jak pro ryby, tak pro chovatele s oslabenou imunitou. U prasat divokých a ptáků (volně žijících i v zajetí) byl zjištěn častý výskyt podmíněně patogenních druhů mykobaktérií. Tato zvířata, jejich trus a okolní prostředí mohou sloužit jako rezervoár mykobakteriálních infekcí pro ostatní zvířata a lidi přicházející do styku s nimi. Automatizovaný MGIT 960 a manuální MGIT tekutý kultivační systém jsou schopny detekovat vyšší procento mykobakterií a v kratším čase než běžně používaná kultivace na pevných médiích. Podařilo se vyvinout PCR systém pro současnou identifikaci všech poddruhů M. avium s použitím tzv. interních amplifikačních standardů pro kontrolu falešně negativních reakcí. Rovněž byl navržen jednoduchý, spolehlivý, rychlý a levný PCR systém pro detekci a diferenciaci jednotlivých poddruhů M. avium z kultur narostlých na pevných kultivačních médiích a z masivně infikovaných živočišných tkání. U prasat pocházejících z různých farem byla zjištěna metodou RFLP nízká genetická variabilita izolátů M. a. avium svědčící o možnosti volného přenosu infekce aviární tuberkulózy mezi jednotlivými farmami. Z publikovaných údajů vyplývá, že formy mykobakterií s deficiencí buněčné stěny mohou, jako produkt interakce infikující baktérie a imunitního systému hostitele, být příčinou chronických onemocnění. Skutečnost, že se tyto formy mykobakterií nedaří z vyšetřovaného materiálu detekovat nutně neznamená, že se jedná o onemocnění zvířat či lidí neinfekčního původu.

Vladimír Beran The submitted thesis represents a summary of publications of the author with comments on individual publications. Conditionally pathogenic mycobacterial species were detected in lymph nodes of pigs fed with the supplementation of both treated and untreated peat. The use of peat as feed supplement for pigs can be risky due to occurrence of mycobacteria. The aquarium environment as well as clinically healthy bred fish contained large quantities of mycobacteria which can pose health risk not only for fish but also for immunocompromised breeders. In wild boar and birds (both in nature and in captivity), frequent occurrence of conditionally pathogenic mycobacterial species have been reported. These animals, their faeces and the surrounding environment might serve as reservoir of mycobacterial infections for other animals and humans. The automated MGIT 960 and manual MGIT liquid culture system are able to detect higher percentage of mycobacteria within a shorter time, compared with commonly used culture on solid media. We have developed PCR system for concurrent identification of all subspecies of M. avium using so called internal amplification standards for checking of false negative reactions. Further, a simple, reliable, fast and inexpensive PCR system for detection and differentiation of individual M. avium subspecies from cultures grown on solid media and from massively infected animal tissues has been designed. Low genetic variability of M. a. avium isolates from pigs originating from different farms has been demonstrated, which is suggestive of a possible free transmission of avian tuberculosis among the farms. The published data show that cell wall deficient forms of mycobacteria can be the cause of chronic disease as a product of interaction between the infecting bacteria and the host’s immune system. The fact that detection of these forms of mycobacteria from the examined materials fails does not necessarily mean that the disease of animals or humans is of non-infectious origin.
