The searching for the evidences of interaction between KATANIN 1 protein and selected mitogen-activated protein kinases

Abstract:

Mitogen aktivované protein kinasy jsou součástí signalizačních drah u rostlin. MAP kinasy fungují v kaskádách. V rámci kaskády přenáší a zesilují signál prostřednictvím fosforylace jednotlivých kinas, které se kaskády účastní. MAPK kaskády se účastní odpovědi na abiotický stres (regulace odpovědi na chlad, sucho, zasolení a oxidační stres) i na biotický stres (součást buněčné imunitní odpovědi). MAPKs jsou také v asociaci s mikrotubuly, kdy se účastní zvětšování buněk, cytokineze, vývoje kořene, kořenových vlásků a epidermálních buněk. S regulací mikrotubulů je také spojen protein katanin. Katanin je heterodimerní protein zkracující mikrotubuly. Tato bakalářská práce se věnovala sledování interakce proteinu katanin s dosud nejlépe prostudovanými MAP kinasami: MPK3, MPK4 a MPK6 účastnícími se mnoha signálních drah. Interakce byla sledována pomocí metody transientní transformace N. benthamiana s využitím bimolekulární fluorescenční komplementace, kde byla potvrzena interakce kataninu s MPK4. Interakce proteinu katanin s MPK3 byla detekována pomocí kvasinkového dvouhybridního experimentu.

Abstract:

Mitogen activated protein kinases are components of signaling pathways in plants. MAP kinases work in cascades. The signa lis transferred and amplified through fosforylation of kinases involved in cascade. MAP kinases are involved in response to abiotic stress (reagulation of response for cold, salinity, drouhg and oxidative stress) and also in response to biotic stress (part of plant immunity). MAPKs are also in association with microtubules, they play role in extending cells, cytokinesis, root development and in development of root hairs and epidermis. Protein katanin is also connected with regulation of microtubules. It's a heterodimeric protein with microtubule severing activity. This bachelor thesis was focused on identificaton of interaction between katanin and the most studied MAP kinases: MPK3, MPK4 and MPK6 involved in many signalling cascades. Interaction was studied by transient tranformation of N. benthamiana and using method, where the interaction of katanin and MPK4 was detected Interaction of katanin and MPK3 was detected by yeast two hybrid experiment.
