Coronaviral proteins with prion-like properties

Abstract:

Priony jsou infekční proteinové částice, které jsou původci především vzácných neurodegenerativních nemocí. Tvorba prionů a tzv. prion-like proteinů je v posledních letech spojována také s neurologickými příznaky objevujících se u pacientů, kteří prodělali onemocnění COVID-19, jehož původcem je koronavirus SARS-CoV-2. Z toho důvodu se do popředí dostává studium prionového potenciálu proteinů nejen koronavirů. Některé proteiny lidských patogenních koronavirů však dosud nebyly prozkoumány. V této práci byl pomocí in silico analýzy predikován prionový potenciál koronavirových proteinů s využitím algoritmu PLAAC. Pro následné potvrzení bylo využito metody třepání pro indukci prionových struktur. Následná detekce byla provedena měřením cirkulárního dichroismu a nativní polyakrylamidovou elektroforézou. Nukleokapsidový protein SARS-CoV, MERS-CoV a SARS-CoV-2 má na základě bioinformatické analýzy nejvyšší pravděpodobnost výskytu prion-like domény. Pro in vitro analýzu byl využit rekombinantní nukleokapsidový protein SARS-CoV-2. Pomocí metody třepání a následné detekce cirkulárním dichroismem se podařilo prokázat změnu v sekundární struktuře proteinu. Naše výsledky však průkazně nepotvrzují tvorbu prionových struktur v nukleokapsidovém proteinu. Nativní elektroforéza naznačila odlišnou migraci třepaných vzorků, ale pro potvrzení je třeba dalších studií. Proto je do budoucna potřeba optimalizovat tuto metodu nebo k potvrzení využít metody jiné. Potvrzení výskytu prion-like domény v nukleokapsidovém proteinu by mohlo v budoucnu pomoct při objasnění neurologických příznaků nejen u pacientů COVID-19. Prionové domény nukleokapsidových proteinů by se tak mohly stát novými cíli pro vývoj terapeutik.

Abstract:

Prions are infectious protein particles that are primarily the cause of rare neurodegenerative diseases. In recent years, the formation of prions and so-called prion-like proteins has also been associated with neurological symptoms appearing in patients who have experienced the disease COVID-19, the causative agent of which is the SARS-CoV-2 coronavirus. For that reason, the study of the prion potential of proteins not only of coronaviruses is coming to the fore. However, some proteins of human pathogenic coronaviruses have not yet been investigated. In this work, the prion potential of coronavirus proteins was predicted using in silico analysis using the PLAAC algorithm. For subsequent confirmation, shaking methods were used to induce prion structures. Subsequent detection was performed by circular dichroism measurement and native polyacrylamide electrophoresis. The nucleocapsid protein of SARS-CoV, MERS-CoV and SARS-CoV-2 based on bioinformatics analysis has the highest probability of the occurrence of a prion-like domain. Recombinant SARS-CoV-2 nucleocapsid protein was used for in vitro analysis. Using the shaking method and subsequent detection by circular dichroism, it was possible to demonstrate a change in the secondary structure of the protein. However, our results do not conclusively confirm the formation of prion structures in the nucleocapsid protein. Native electrophoresis indicated a different migration of shaken samples, but further studies are needed to confirm. Therefore, in the future, it is necessary to optimize this method or to use other methods for confirmation. Confirmation of the occurrence of a prion-like domain in the nucleocapsid protein could in the future help clarify neurological symptoms not only in patients with COVID-19. Prion domains of nucleocapsid proteins could thus become new targets for the development of therapeutics.
