Detection of proteins related to induced differentiation of sarcoma cell lines

Abstract:

Nádorová onemocnění jsou hned za úrazy druhou nejčastější příčinou dětských úmrtí u dětí. Ačkoli v posledních desetiletích nastaly pokroky v léčbě dětských nádorových onemocnění, jsou výsledky u dětí s některými typy nádorů, např. metastatickými sarkomy stále nedostačující. Biologická terapie založená na indukci diferenciace buněk do terminálního stadia se používá v různých léčebných protokolech, například v protokolu metronomické léčby COMBAT. V rámci této práce byla sledována exprese osteogenních (alkalická fosfatáza, osteopontin) a myogenních (vimentin, desmin, tropomyosin) markerů u osteosarkomových a rabdomyosarkomových buněk pomocí imunoblottingu. Jako induktor diferenciace byla použita kyselina all-trans retinová (ATRA), a její účinky byly modulovány pomocí inhibitorů 5-lipoxygenázy a cyklooxygenázy-2, konkrétně kyseliny kávové a celecoxibu. Byla zjištěna stimulace hladiny diferenciačních markerů po ovlivnění ATRA ve všech sledovaných liniích, a modulaci této stimulace za použití kombinace inhibitorů a ATRA u linií Saos-2, OSA-01 a NSTS-08. Získané výsledky demonstrují schopnost sledovaných inhibitorů potencovat diferenciační účinek ATRA, ačkoli tyto efekty, jako i časový průběh diferenciace jsou specifické pro jednotlivé buněčné linie.

Abstract:

Malignant diseases of children are the second leading cause of childhood deaths after injuries. Despite new approaches in the field of pediatric oncology in recent decades remains the treatment of some cancers such as metastatic sarcomas still insufficient. Biological therapy based on the induction of cell differentiation into the mature stadium is used in some therapeutic approaches, e.g. in metronomic protocol COMBAT. The expression of osteogenic (alkaline phosphatase, osteopontin) and myogenic (vimentin, desmin, tropomyosin) differentiation markers was evaluated in osteosarcoma and rhabdomyosarcoma cell lines using immunoblotting. As an inducer of cell differentiation all-trans retinoic acid (ATRA) was used, and its effects were modulated using inhibitors of 5-lipoxygenase and cyclooxygenase-2, caffeic acid and celecoxib. Increased levels of differentiation markers were observed after treatment with ATRA in all studied cell lines, as also the modulation of this stimulation after combined treatment (ATRA and inhibitors) in Saos-2, OSA-01 and NSTS-08 cell lines. The obtained results demonstrate the capability of LOX/COX inhibitors to potentiate the differentiation effect of ATRA in sarcoma cell lines, although these effects, as well as the time course of differentiation events are cell-line specific.
