Abstract:

Využití biotechnologií je nadějí pro budoucnost. Použití bakterií a jejich enzymů je jednou z oblastí, jak při nižších nákladech dosáhnout vyšší efektivity v mnoha oblastech lidského počínání. Bakterie Acidithiobacillus ferrooxidans hraje značnou roli při operacích provázejících těžbu či zpracování kovů, ať již v pozitivním (biohydrometalurgie) či negativním (tvorba kyselých důlních vod) smyslu. Ačkoliv se jejím metabolismem zabývala již řada pracovišť, stále zůstávají mnohé otazníky, zejména v oblasti metabolismu sloučenin síry. Jednou z cest, jak zkoumat jednotlivé enzymy zapojené do metabolismu síry, je jejich příprava v rekombinantní podobě a následné studium těchto proteinů. V rámci této práce byla pozornost zaměřena na skupinu enzymů thiosulfát:kyanidsulfurtransferas (rhodanas). V genomu se nachází osm genů pro potenciální rhodanasy. Byla ověřena možnost amplifikace sedmi těchto genů pomocí PCR. Pro další studium byla vybrána jedna potenciální rhodanasa (RHC), která dosud nebyla detailněji zkoumána v žádné publikované práci. Jí odpovídající gen byl naklonován do plasmidu a vložen do buněk Escherichia coli. Po optimalizaci použitého kmenu E. coli, teploty a doby exprese bylo dosaženo nadprodukce rekombinantního enzymu. Takto získaný protein byl následně izolován a přečištěn z inkluzí pomocí chromatografie (FPLC). Provedený renaturační krok vedl k enzymu s rhodanasovou aktivitou. Pro nalezení biologické funkce tohoto enzymu jsou nutné další kroky. Bylo by vhodné optimalizovat renaturaci za účelem zisku maximálního množství aktivního proteinu, u něj následně stanovit rhodanasovou aktivitu a případné další parametry (různé substráty, inhibice, teplotní a pH optima,…). K pochopení jeho významu by rovněž přispěla případná identifikace tohoto proteinu na proteinové mapě získané 2D elektroforezou proteinů z Acidithiobacillus ferrooxidans rostoucích na různých substrátech. Výhodné by bylo rovněž připravit krystal proteinu a určit jeho detailní strukturu.

Abstract:

Use of biotechnologies is a hope for the future. Use of bacteria and their enzymes is one of the fields, where we can obtain higher efficiency with lower costs. The bacterium Acidithiobacillus ferrooxidans plays an important role in processes connected with mining and metal processing in positive (biohydrometallurgy) and also negative (acid mine drainage) way. Despite many teams has studied the metabolism of this bacterium, there are still many questions, especially about sulphur compounds metabolism. One of the ways, how to study single enzymes involved in sulphur metabolism, is their preparation in recombinant form and study of these recombinants. In this work we aimed at rhodaneses (thiosulphate:cyanide sulfurtransferases). There are eight genes for these enzymes in the genome. We have verified the possibility of amplification of these genes via PCR. For next study we have chosen one of the potential rhodaneses (RHC), which has not been studied so far. Its gene has been cloned into plasmid and put into E. coli cells. After the optimization of the strain, temperature and time of expression we achieved overexpression of recombinant enzyme. This protein has been isolated and purified. This process has lead to enzyme with rhodanese activity. There are next steps necessary to find the biological function of this enzyme. It would be useful to optimize renaturation to gain as much active protein as possible. Than determine activity and other parameters (substrates, inhibitions, temperature and pH optima, etc.) To understand its role in organism we can also try to identify this protein in 2D electrophoresis maps of Acidithiobacillus ferrooxidans grown on different substrates. Preparation of crystal and determination of protein structure would be also desirable.
