Cyanobacterial neurotoxins

Abstract:

Problematika sinic a s tím spojená tvorba sinicových vodních květů patří v posledních letech k velmi diskutovaným tématům. Měnící se přírodní podmínky a intenzivnější antropogenní činnost umožňují sinicím obývat oblasti, kde se dříve vůbec nevyskytovaly. Nebezpečnost sinic tkví v jejich schopnosti produkovat sekundární metabolity s toxickým potenciálem, které pak mohou negativně ovlivnit organismy nacházející se v jejich blízkosti. V posledních letech se výzkum zaměřuje na zjišťování mechanismu působení toxických látek produkovaných sinicemi na jednotlivé orgánové soustavy, jako je například vliv látek na nervovou soustavu. Nicméně většina studií se zabývá spíše jednotlivými látkami než směsí látek, které se v prostředí vyskytují nejčastěji. Tyto látky mohou navzájem interagovat a jejich účinky mohou být jiné, než jako tomu bylo u samostatné látky. Tato diplomová práce se zaměřuje na neurotoxický potenciál dvou sinicových extraktů odebraných z prostředí v porovnání se standardním toxikantem kyselinou all-trans retinovou, která se přirozeně vyskytuje v sinicových exudátech a je znám její teratogenní efekt na raná vývojová stádia ryb Danio rerio. Měřeným parametrem je změna pohybové aktivity embryí. Použitou metodou pro měření pohybové aktivity embryí je light/dark test. Měnící se světelné podmínky vedou ke změně aktivity embryí, což odráží přirozené chování embryí Danio rerio. Ve vzorcích sinicové biomasy, které byly odebrány z vodní nádrže Vranov a z rybníku Ruda, byly naměřeny retinoidní látky (all-trans retinová kyselina, 9-cis retinová kyselina, retinal, 4-keto all-trans retinová kyselina, 4-keto retinal), které také mohou mít vliv na nervový systém. Z toxinů byl pouze microcystin nad limitem detekce, ale jen u vzorku z vodní nádrže Vranov obsahující sinice druhu Microcystis aeruginosa. Výsledky ukazují, že ke změně v pohybové aktivitě mezi kontrolními a exponovanými embryi dochází jak u expozice extrakty z prostředí, tak při expozici standardní látce kyselině all-trans retinové, nicméně tyto změny nejsou statisticky významné. Pouze vzorek z vodní nádrže Vranov způsobil při 3hodinové expozici při koncentraci 0,2 g dw/L výrazné zvýšení pohybové aktivity embryí Danio rerio, a to ve světelné i temné fázi. Hypotéza o působení extraktu sinicové biomasy na nervový systém Danio rerio však potvrzena nebyla.

Abstract:

The issue of cyanobacteria and the associated creation of cyanobacterial water blooms belong to the discussed topic in recent years. Changing natural conditions and more intense anthropogenic activity allow cyanobacteria to inhabit areas where they did not exist previously. The danger of cyanobacteria is in their ability to produce secondary metabolites with toxic potential, which can then negatively influence organisms in their proximity. In recent years, research has focused on detecting the mechanism of action of toxic substances produced by cyanobacteria on individual organ systems, such as the effects of substances on the nervous system. However, most studies deal with substances rather than mixtures of substances which most occur in the environment. These substances can interact with each other and their effects may be different from that of the separate substance. This diploma thesis focuses on the neurotoxic potential of two cyanobacterial extracts extracted from the environment in comparison with the standard all-trans retinoic acid toxicant that naturally occurs in cyanobacterial exudates and it is known its teratogenic effect on the early stages of Danio rerio. The measured parameter is the change in the embryo's movement activity. The method used to measure embryo's physical activity is the light/dark test. Changing light conditions lead to a change in embryo activity, what reflects the natural behavior of the Danio rerio embryos. The retinoid substances (all-trans retinoic acid, 9-cis retinoic acid, retinal, 4-keto all trans retinoic acid, 4-keto retinal) were measured in the cyanobacterial biomass samples taken from the Vranov reservoir and from the Ruda pond, which can also affect the nervous system. From the toxins, only the microcystin was above the detection limit, but only in sample from the Vranov reservoir with cyanobacterium Microcystis aeruginosa. The results show changes in locomotor activity between control and exposed embryos by extracts from the environment and standard toxicant all-trans retinoic acid, but these changes are statistically insignificant. Only a sample from Vranov reservoir 3-hours exposure at concentration 0,2 g dw/L caused a significant increase of the Danio rerio embryo´s movement activity in light phase, also in dark phase. The hypothesis of the effect of the extract of cyanobacteria on the nervous system Danio rerio was not confirmed.
